中文摘要: 利用Lacticaseibacillus paracasei PC18发酵黑莓原浆,对产品加工过程中营养化学物质含量及抗氧化活性进行连续测定,并采用超高效液相-质谱联用(ultra-high performance liquid chromatography-mass spectrometry,UPLC-MS)技术对发酵前后酚类成分定性和定量分析。结果表明,黑莓发酵液在发酵0~24 h间还原糖含量由51.79 mg/mL显著增加至66.65 mg/mL(P<0.05),随后呈波动性降低;总酚含量呈现逐渐下降的趋势,经过48 h发酵后,由1 006.81 μg/mL下降至871.19 μg/mL后维持相对稳定;黑莓发酵液的抗氧化活性在0~48 h由3.82 mmol TEAC/L显著增加至4.65 mmol TEAC/L(P<0.05),于发酵72 h时达到最高,为5.13 mmol TEAC/L。经过48 h发酵,黑莓发酵液中共鉴定出13种差异变化的酚酸化合物,其中12种酚酸含量增加,仅新绿原酸含量下降,其中香草酸、原儿茶酸、龙胆酸和对羟基苯甲酸含量的增幅最大,依次为85.07%、72.79%、29.33%和28.97%。相关性分析表明,L. paracasei PC18应用于黑莓原浆发酵,能够促进黑莓酚类物质的生物转化,从而为黑莓产品发挥更高抗氧化活性提供丰富的物质基础。
Abstract:The fermentation of blackberry pulp using Lacticaseibacillus paracasei PC18 was conducted, with continuous measurement of the nutritional chemical content and antioxidant activity throughout the processing.Ultra-high performance liquid chromatography-mass spectrometry (UPLC-MS) was used for the qualitative and quantitative analysis of polyphenolic compounds before and after fermentation. The results showed that the reducing sugar content of the blackberry fermentation liquid increased significantly from 51.79 mg/mL to 66.65 mg/mL after 24 h fermentation, and then fluctuated downwards. The total phenolic content gradually decreased, stabilizing after 48 h, from 1 006.81 μg/mL to 871.19 μg/mL(P<0.05). The antioxidant activity of the blackberry fermentation liquid increased from 3.82 mmol TEAC/L to 4.65 mmol TEAC/L over the first 48 h(P<0.05), reaching a peak of 5.13 mmol TEAC/L at 72 h. After 48 h fermentation, 13 polyphenolic compounds with significant changes were identified, with 12 showing an increase in concentration, except for chlorogenic acid, which decreased. The largest increases were observed in vanillic acid, protocatechuic acid, gentisic acid, and p-hydroxybenzoic acid, with increases of 85.07%, 72.79%, 29.33%, and 28.97%, respectively. Correlation analysis indicated that the fermentation of blackberry pulp with L. paracasei PC18 promotes the biotransformation of phenolic compounds in the blackberry, providing a rich material basis for enhancing the antioxidant activity of blackberry products.
