中文摘要: 为探究不同提取方式对长茎葡萄蕨藻多糖及体外抗氧化活性的影响,分别以挤压提取、水浴提取及碱提取3种方法依次分级提取长茎葡萄蕨藻多糖,比较通过不同提取方式分级提取的多糖提取率、单糖组成和相对分子质量;通过测定DPPH自由基、ABTS+自由基和羟基自由基清除能力考察其抗氧化活性。结果 表明:不同提取方式多糖得率由高到低依次为碱提取、水浴提取、挤压提取,所获得的多糖提取率分别为28.83%、5.30%和3.85%,其中挤压提取和水浴提取所获得的多糖单糖组成相似,主要为半乳糖、葡萄糖、木糖和甘露糖,碱提法所得多糖其单糖组成为木糖,经鉴定为β-1,3-木聚糖;不同提取方式所得多糖其抗氧化活性也有一定差异,碱提取得到的多糖对DPPH自由基和羟基自由基的清除能力最强,尤其对羟基自由基的清除能力尤为明显,高达97.60%;水浴提取得到的多糖ABTS+自由基清除能力最强,碱提取次之,挤压提取最弱。经对比发现,β-1,3-木聚糖显示出最佳的抗氧化活性,并呈浓度依赖性。
Abstract:To investigate the effects of different extraction methods on the polysaccharides of Caulerpa lentillifera and their in vitro antioxidant activities,the polysaccharides were successively extracted by extrusion extraction,water bath extraction and alkali extraction. The extraction rates,monosaccharide compositions and relative molecular masses of the polysaccharides obtained by different extraction methods were compared. The antioxidant activities were investigated by measuring the scavenging abilities of DPPH radicals,free radicals and hydroxyl radicals. The results showed that the extraction rates of polysaccharides obtained by different extraction methods were in the order of alkali extraction>water bath extraction>extrusion extraction,which were 28.83%,5.30% and 3.85%,respectively. The monosaccharide compositions of the polysaccharides obtained by extrusion extraction and water bath extraction were similar,mainly consisting of galactose,glucose,xylose and mannose. The monosaccharide composition of the polysaccharides obtained by alkali extraction was xylose,which was identified as β-1,3-xyloglucan. The antioxidant activities of the polysaccharides obtained by different extraction methods also showed certain differences. The polysaccharides obtained by alkali extraction had the strongest scavenging abilities for DPPH radicals and hydroxyl radicals,especially for hydroxyl radicals,reaching up to 97.60%. The polysaccharides obtained by water bath extraction had the strongest scavenging ability for free radicals,followed by alkali extraction and extrusion extraction. It was found that β-1,3-xyloglucan showed the best antioxidant activity and presented a concentration-dependent manner.
