中文摘要: 提高机体免疫力对促进肌肉骨骼系统发育及预防骨质疏松具有重要意义。该研究在钙制剂和维生素D3的基础上,通过添加乳清蛋白和亮氨酸复配得到一款肌肉骨骼系统膳食营养补充剂,并探究其对小鼠免疫功能的影响。将160只ICR小鼠随机分为空白组和低、中、高剂量组,连续灌胃30 d后,对小鼠免疫脏器与体质量之比、细胞免疫、体液免疫和血清细胞因子水平进行评价。结果 显示,与空白组相比,灌胃中剂量补充剂能显著增加胸腺指数,低、中剂量显著提升抗体生成能力和血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)浓度(P<0.05);而中、高剂量对小鼠的耳肿胀率、巨噬细胞吞噬率、自然杀伤(natural killer,NK)细胞活性、白细胞介素-6(interleukin-6,IL-6)浓度有显著提高作用(P<0.05、P<0.01);此外,各组血清白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-10(interleukin-10,IL-10)浓度无显著变化(P>0.05),但灌胃中剂量能显著提升空斑数、碳廓清吞噬指数(P<0.05)。上述结果提示,该营养补充剂具有增强细胞免疫和体液免疫的功效,中剂量(3.8 g/kg体质量)即可产生明显效果。
Abstract:Improving the body′s immunity is of great significance for promoting musculoskeletal system development and preventing osteoporosis. In this study,a musculoskeletal system dietary supplement was formulated based on calcium and vitamin D3,with the addition of whey protein and leucine,to explore its effect on immune function in mice. A total of 160 ICR mice were randomly divided into a blank group and low,medium,and high-dose groups. After 30 days of continuous gavage,the immune organ/body weight ratio,cellular immunity,humoral immunity,and serum cytokine levels were evaluated. The results showed that compared with the blank group,gavage with low and medium doses of the supplement significantly increased the thymic index,low and medium doses of the supplement significantly increased antibody production capacity,and serum tumor necrosis factor-α (TNF-α) concentration (P<0.05). Furthermore,medium and high doses significantly enhanced the ear swelling rate,macrophage phagocytosis rate,natural killer (NK) cell activity,and serum interleukin-6 (IL-6) concentration in mice (P<0.05,P<0.01). Additionally,there were no significant changes in serum interleukin-1β (IL-1β) and interleukin-10 (IL-10) concentrations across all groups(P>0.05),but gavage with the medium dose significantly increased the number of empty spots and the carbon contour phagocytosis index (P<0.05). These results suggested that the nutritional supplement had the effect of enhancing both cellular and humoral immunity,with the medium dose (3.8 g/kg BW) showing significant effects.
