中文摘要: 以葛根、石斛、薏苡仁、陈皮、枳椇子、菊花、甘草、薄荷、山楂等药食同源物质为主要原料,开发一种具有护肝作用的饮料,并利用小鼠急性酒精性肝损伤模型,评估石斛葛根饮的肝脏保护作用。结果 表明,石斛葛根饮能够提高急性酒精性肝损伤小鼠体质量,降低肝脏指数,还能缓解小鼠肝细胞空泡化和核分裂增多等状况;同时,它可降低小鼠肝组织谷草转氨酶(aspartate aminotransferase,AST)、谷丙转氨酶(alanine aminotransferase,ALT)酶活性,减少低密度脂蛋白(low-density lipoprotein,LDL)、甘油三酯(triglycerides,TG)、胆固醇(cholesterol,CH)和丙二醛(malondialdehyde,MDA)含量,提高超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)含量。肠道菌群分析表明,石斛葛根饮能够改善50%乙醇引起的小鼠肠道菌群纷乱,提高伯克霍尔德菌、螺菌、疣微菌、乳杆菌、丹毒丝菌丰度。综上,石斛葛根饮能够改善50%乙醇诱导的肝损伤,具有开发成为护肝健康食品的潜力。
Abstract:A Shihu Gegen beverage with hepatoprotective effect was developed with Puerariae lobatae Radix(Gegen),Dendrobii officinalis Caulis (Shihu),Coicis Semen,Citri reticulatae Pericarpium,Hoveniae Semen,Chrysanthemi morifolit Flos,Glycyrrhizae Radix et Rhizoma,Menthae haplocalycis Herba,and Crataegi Fructus as the main raw materials. Its hepatoprotective effect was investigated via a mouse model of acute alcoholic liver injury. The results demonstrated that the Shihu Gegen beverage increased the body mass,reduced the liver index,and alleviated hepatocyte vacuolation and excessive nuclear division in the liver of the mouse model. Furthermore,the beverage decreased the activities of aspartate aminotransferase and alanine aminotransferase,lowered the levels of low-density lipoprotein,triglycerides,cholesterol,and malondialdehyde,and enhanced the levels of superoxide dismutase and glutathione peroxidase in the liver tissue. Analysis of the gut microbiota revealed that the beverage ameliorated the gut microbiota dysbiosis caused by 50% ethanol exposure,improving the abundance of Burkholderia,Spirillum,Verrucomicrobium,Lactobacillus,and Erysipelothrix.These findings suggested that the Shihu Gegen beverage had the potential to be developed into a functional health food product for alleviating 50% ethanol-induced liver injury.
