中文摘要: 该研究以牡蛎肽为原料,通过细胞毒性检测试剂盒检测细胞活力,确认牡蛎肽无毒性作用。建立RAW264.7细胞炎症模型,测定RAW264.7细胞一氧化氮含量,运用酶联免疫吸附测定法检测白介素-1β( interleukin-1β,IL-1β)促炎因子的分泌量,再通过实时荧光定量聚合酶链式反应(quantitative polymerase chain reaction,qPCR)检测单核细胞趋化蛋白-1(monocyte chemotactic protein,MCP-1)、干扰素-β(interferon-β,IFN-β)、调节激活正常T-细胞表达分泌因子(recombinant regulated on activation in normal T-cell expressed and secreted,RANTES)、白介素-6( interleukin-6,IL-6)、巨噬细胞炎性蛋白-2( macrophage inflammatory protein-2,MIP-2)、巨噬细胞炎性蛋白-1α( macrophage inflammatory protein-1α,MIP-1α)细胞因子信使核糖核酸(messenger RNA,mRNA)的表达水平,探究牡蛎肽对相关炎症因子的影响;通过免疫印记法检测炎症信号通路蛋白核因子-κB p65(nuclear factor-k-gene binding p65,NF-κB p65)、髓样分化因子88( recombinant myeloid differentiation factor 88,MYD88)、NLR家族Pyrin域蛋白3 (NLR family,pyrin domain containing protein 3,NLRP3)、泛素结合蛋白62(ubiquitin binding protein 62,p62)的表达量。研究结果显示,牡蛎肽可通过调控NF-κB炎症信号通路和炎症小体NLRP3,从而降低IL-1β、MCP-1、IFN-β、RANTES、IL-6、MIP-2、MIP-1α 的表达来发挥抗炎作用。

Abstract:This study uses oyster peptides as raw materials. Cell viability was detected using the cytotoxicity assay kit to confirm that oyster peptides had no toxic effect. An inflammation model of RAW264.7 cells was established,and the content of nitric oxide in RAW264.7 cells was determined. The secretion of the pro -inflammatory factor interleukin-1β( IL-1β) was detected by enzyme-linked immunosorbent assay,and the messenger RNA( mRNA) expression levels of cytokines,including monocyte chemotactic protein-1(MCP-1),interferon-β( IFN-β),recombinant regulated on activation in normal T-cell expressed and secreted( RANTES),interleukin-6( IL-6),macrophage inflammatory protein-2( MIP-2),and macrophage inflammatory protein-1α(MIP-1α) detected by real-time fluorescence quantitative polymerase chain reaction( qPCR) to explore the effect of oyster peptides on related inflammatory factors. The expressions of inflammatory signaling pathway protein nuclear factor-k-gene binding p65 (NF-κB p65),recombinant myeloid differentiation factor 88(MYD88),NLR family,pyrin domain containing protein 3( NLRP3),and ubiquitin binding protein 62( p62)were detected by western blotting. The results showed that oyster peptides may exert anti-inflammatory effects by regulating the MYD88/NF-κB inflammatory signaling pathway and inflammasome NLRP3,thereby reducing the expressions of IL-1β,MCP-1,TGF-β,RANTES,IL-6,MIP-2,and MIP-1α .
