中文摘要: 为阐明温度波动幅度对生鲜肉品质和宰后能量代谢酶磷酸化的影响,以猪背最长肌为材料,分析对照组(4 ℃恒温)、波动组Ⅰ(4 ℃贮藏3 h→7 ℃贮藏3 h→4 ℃贮藏3 h)和波动组Ⅱ(4 ℃贮藏3 h→15 ℃贮藏3 h→4 ℃贮藏3 h)在贮藏过程中肉品质(pH值、蒸煮损失、剪切力)、糖原含量、腺嘌呤核苷三磷酸(adenosine triphosphate,ATP)含量、乳酸含量、糖原磷酸化酶(glycogen phosphorylase,PYGM)、二磷酸果糖醛缩酶A(aldolase A,ALDOA)和磷酸丙糖异构酶(triosephosphate isomerase,TPI)等能量代谢酶活性及其磷酸化水平的变化。结果 表明:波动组Ⅱ样本的pH值在宰后120 h显著高于对照组,蒸煮损失在宰后72 h显著高于对照组,剪切力在宰后72 h和120 h显著高于对照组(P<0.05)。波动组Ⅱ样本的糖原含量在宰后12 h显著低于对照组,乳酸含量在宰后24 h显著高于对照组(P<0.05),PYGM和TPI的活性和磷酸化水平在宰后24 h均显著低于对照组(P<0.05)。不同处理组的ALDOA活性和磷酸化水平差异不显著(P>0.05)。综上,温度波动幅度增大提升了磷酸化对PYGM活性的促进作用和对TPI活性的抑制作用,加快宰后能量代谢,促使肌肉pH值下降,生鲜肉保水性下降。
Abstract:The effects of temperature fluctuation amplitude on the quality of fresh meat and the phosphorylation of energy metabolism enzymes after slaughter were studied. The longissimus dorsi muscle samples of pork were assigned into three groups:control (4 ℃ constant temperature),fluctuation I (4 ℃ storage for 3 h→7 ℃ storage for 3 h→4 ℃ storage for 3 h),and fluctuation II (4 ℃ storage for 3 h→15 ℃ storage for 3 h→4 ℃ storage for 3 h). The three groups were compared regarding the meat quality (pH value,cooking loss,and shear force),content of glycogen,ATP,and lactic acid,and activities and phosphorylation levels of glycogen phosphorylase(PYGM),aldolase A(ALDOA),and triosephosphate isomerase(TPI) during storage. The results showed that the fluctuation II group had higher pH 120 h after slaughter,higher cooking loss 72 h after slaughter,and higher shear force 72 h and 120 h after slaughter than the control group(P<0.05). The fluctuation II group had lower glycogen content 12 h after slaughter and higher lactate content 24 h after slaughter than the control group (P<0.05). The activities and phosphorylation levels of PYGM and TPI in the fluctuation II group were lower than those in the control group 24 h after slaughter (P<0.05). There was no significant difference in the activity or phosphorylation level of ALDOA among different groups(P>0.05). In conclusion,the increase in temperature fluctuation amplitude exacerbated the promoting effect of phosphorylation on PYGM activity and the inhibitory effect on TPI activity,accelerated energy metabolism after slaughter,promoted pH decrease,and reduced the water retention of fresh meat.
