中文摘要: 该研究聚焦于蔬菜发酵专用乳酸菌的筛选鉴定以及直投式发酵剂的制备。通过体外试验初步筛选得到耐盐及产酸的候选菌株,将候选菌株接种到雪菜发酵体系中,通过分析雪菜发酵期间的理化特性(pH值、可滴定酸含量、盐度、亚硝酸盐含量、菌落总数)、挥发性风味特征以及感官特性,并结合16S rDNA技术,鉴定得到具备优良蔬菜发酵特性的乳酸菌菌株。制备直投式发酵剂,并研究该发酵剂的贮藏稳定性。结果表明,从141株菌中筛选得到10株具有耐盐和产酸的候选菌株,将10株候选菌株接种到雪菜发酵体系中,发现菌株WZW-1在理化特性、挥发性风味物质种类和含量上均优于其他菌株,感官评价综合得分最高,经鉴定命名为植物乳植杆菌WZW-1。以优势菌株WZW-1制备的直投式发酵剂冻干存活率为(97.58±0.83)%,活菌数为11.82 lg(CFU/g),且在-20 ℃下贮藏1年后活菌数仍可维持在10.12 lg(CFU/g)。
Abstract:This study focused on the screening and identification of lactic acid bacteria specific for vegetable fermentation and the preparation of a direct vat set starter.Salt-tolerant and acid-producing candidate strains were preliminarily obtained through in vitro screening.The candidate strains were inoculated into the potherb mustard fermentation system.The physicochemical properties(pH value,titratable acid content,salinity,nitrite content,and aerobic plate count),volatile flavor characteristics,and sensory attributes during fermentation were analyzed in combination with 16S rDNA technology.Lactic acid bacteria strains with excellent characteristics for vegetable fermentation were identified.A direct vat set starter was prepared,and its storage stability was investigated.The results showed that ten candidate strains with salt tolerance and acid-producing ability were screened from 141 strains.These ten candidate strains were inoculated into the potherb mustard fermentation system,and strain WZW-1 exhibited superior performance in physicochemical properties,types,and contents of volatile flavor compounds,and received the highest overall score in sensory evaluation.The strain was identified as Lactobacillus plantarum WZW-1.The freeze-drying survival rate of the direct vat set starter prepared with the dominant strain WZW-1 was(97.58±0.83)%,with an aerobic plate count of 11.82 lg(CFU/g).After one year of storage at -20 ℃,the aerobic plate count remained at 10.12 lg(CFU/g).
