中文摘要: 通过体外模拟胃肠液环境分析植物乳植杆菌SLlac-23的耐受性,结合急性/慢性经口毒性实验、抗生素敏感性试验及血琼脂平板法溶血活性检测,综合评价菌株生物安全性。进一步通过检测小鼠模型血清谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、肌酐(creatinine,Cr)和尿素氮(blood urea nitrogen,BUN)等肝肾功能指标,验证长期摄入对宿主的影响。结果表明:SLlac-23在模拟胃液和肠液中存活率均在88%以上,表现出良好的胃肠耐受性,且该菌株不具备溶血活性;经口毒性实验可知无小鼠死亡或异常行为;与对照组相比,小鼠体质量、脏器指数及肝肾功能指标均无显著差异(P>0.05)。基于以上分析,具有降尿酸功效的植物乳植杆菌SLlac-23具备良好的安全性及在临床上开发为降尿酸益生菌制剂的潜力。
Abstract:The biosafety of Lactobacillus plantarum SLlac-23 was comprehensively evaluated by analyzing its tolerance in simulated gastric and intestinal fluid environments in vitro,combined with acute and chronic oral toxicity tests,antibiotic susceptibility tests,and hemolytic activity detection using the blood agar plate method.Additionally,the impact of long-term intake on host health was verified by measuring hepatic and renal function indicators in a mouse model,including serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),creatinine(Cr),and blood urea nitrogen(BUN).The results showed that the survival rate of SLlac-23 exceeded 88% in both simulated gastric and intestinal fluids,indicating good gastrointestinal tolerance.The strain exhibited no hemolytic activity.No mortality or abnormal behavior was observed in oral toxicity tests.Compared with the control group,there were no significant differences in body weight,organ indices,or hepatic and renal function indicators(P>0.05).Based on these findings,Lactobacillus plantarum SLlac-23 with the uric acid-lowering effect exhibited excellent safety and showed potential for clinical development as a probiotic preparation for reducing uric acid levels.
