中文摘要: 为优化多花黄精多糖提取工艺,以多糖提取率为指标,通过单因素分析和响应面法,优化酶辅助提取多花黄精多糖工艺。结果表明,多花黄精多糖的最佳提取工艺条件为料液比1∶18( g/mL)、复合酶添加量1.0%、纤维素酶与木瓜蛋白酶质量比3∶7、酶解温度53 ℃、酶解时间50 min,在此条件下,多糖提取率达20.89%。该提取液多糖浓度为50 mg/mL时,对DPPH自由基清除率为81.39%,IC50为21.12 mg/mL。利用该提取液制备的黄精艾尔精酿啤酒,符合国家相关标准并具有黄精特有香气和口感。
Abstract:To optimize the polysaccharide extraction from Polygonatum cyrtonema Hua,this paper aimed to optimize the enzyme-assisted extraction process using single-factor analysis and response surface methodology,with the polysaccharide extraction rate as the evaluation indicator. The results showed that the optimal extraction process conditions for polysaccharide were a solid-liquid ratio of 1∶18(g/mL),enzyme addition of 1.0%,a mass ratio of cellulase to papain of 3∶7,an enzymatic hydrolysis temperature of 53 ℃,and an enzymatic hydrolysis time of 50 min. Under these conditions,the extraction rate reached 20.89%. When the polysaccharide concentration of the extract was 50 mg/mL,the DPPH radical scavenging rate was 81.39%,with an IC50 value of 21.12 mg/mL. The Huangjing ale beer prepared using the aqueous extract of Polygonatum cyrtonema Hua meets national standards and possesses the unique aroma and flavor of Polygonatum.
