中文摘要: 为获取金枪鱼血肽的最适蛋白酶并优化最佳酶解制备工艺,该研究利用5种蛋白酶酶解金枪鱼鱼血,以水解率为考察指标筛选出最适蛋白酶,并通过单因素试验结合响应面分析法确定最佳酶解工艺,同时分析该工艺对金枪鱼鱼血氨基酸含量的影响。金枪鱼鱼血中蛋白质含量为(74.03±0.99) g/100 g,脂肪含量为(0.73±0.03) g/100 g,灰分含量为(9.03±0.04) g/100 g。选用风味蛋白酶作为酶解最优酶,金枪鱼血肽最佳酶解工艺为酶解时间230 min、酶解温度65 ℃和酶解pH值为8.8,在此条件下蛋白水解率为21.32%,与响应面预测值拟合较好。另外,金枪鱼血肽必需氨基酸含量丰富,与联合国粮食及农业组织/世界卫生组织建议的必需氨基酸模式标准基本一致。
Abstract:To obtain the optimal protease for tuna blood peptides and to optimize the enzymatic preparation process,five proteases were used to hydrolyze tuna blood,and the optimal protease was selected based on the degree of hydrolysis. The optimal enzymatic hydrolysis conditions were determined via single-factor experiments combined with response surface methodology,and the effect of the process on the amino acid content of tuna blood was analyzed. The tuna blood contained (74.03±0.99) g/100 g protein,(0.73±0.03) g/100 g fat,and(9.03±0.04) g/100 g ash. Flavor protease was selected as the most suitable enzyme. The optimal enzymatic hydrolysis process was 230 minutes at 65 ℃ and pH of 8.8,under which the degree of hydrolysis reached 21.32%,closely matching the predicted value. Furthermore,the content of essential amino acids in the tuna blood peptides were abundant and generally consistent with the essential amino acid pattern standards recommended by FAO/WHO.
