嗜酸乳杆菌发酵黑木耳饮料的工艺优化及产品活性成分测定

中文摘要: 黑木耳营养丰富,含有多种活性成分,黑木耳中很多大分子物质不能被人体吸收,利用发酵的方式将大分子营养物质分解成更易吸收的小分子成分。该研究以黑木耳为原料,采用嗜酸乳杆菌对其进行发酵,以感官评分为指标,在单因素试验的基础上,采用响应面法对嗜酸乳杆菌发酵工艺进行优化,并通过液相色谱、硫酸蒽酮、考马斯亮蓝法分析黑木耳发酵饮料有机酸、多糖、蛋白质的含量。确定嗜酸乳杆菌发酵黑木耳饮料的最佳工艺条件为液固比为80∶1(mL/g)、接种量为1.5%、低聚果糖添加量为2.0%、发酵时间为24 h。在此条件下,总酸含量5.62 g/kg,多糖含量6.85 g/L,蛋白质含量2.65 g/L。未发酵饮料中仅检测出草酸一种有机酸,而嗜酸乳杆菌发酵黑木耳饮料中检测出以乳酸和琥珀酸为主的4种有机酸。在发酵的过程中,产生乳酸、琥珀酸与柠檬酸,且草酸含量增加。

Abstract:Auricularia auricula is rich in nutrition and contains various active ingredients. Many macromolecular substances in Auricularia auricula cannot be absorbed or utilized by the human body. Fermentation can decompose macromolecular nutrients into components of smaller molecules that are more easily absorbed. In this study,Auricularia auricula was taken as the raw material,which was fermented by Lactobacillus acidophilus.Then,with sensory score as the index,the response surface method was used to optimize the fermentation process of Lactobacillus acidophilus based on the single factor experiment. Additionally,the content of organic acids,polysaccharides,and proteins in the fermented Auricularia auricula beverage was analyzed by methods such as liquid chromatography,the anthrone-sulfuric acid method,and the coomassie brilliant blue method.The optimal technological conditions for Auricularia auricula beverage fermented by Lactobacillus acidophilus were determined as follows:a liquid-solid ratio of 80∶1 (mL/g),a starter inoculation amount of 1.5%,a fructooligosaccharide addition amount of 2.0%,and a fermentation time of 24 h. Under these conditions,the total acid content was 5.62 g/kg,the polysaccharide content was 6.85 g/L,and the protein content was 2.65 g/L. Oxalic acid was the only organic acid that could be detected in the unfermented Auricularia auricula beverage,while four kinds of organic acids,mainly lactic acid and succinic acid,could be detected in the beverage fermented by Lactobacillus acidophilus. During the fermentation process,lactic acid,succinic acid,and citric acid were produced,while the oxalic acid content was increased.
