中文摘要: 为研究乙醇体积分数对陕北狗头枣可食用部分(枣皮和枣肉)总多酚提取效果的影响,该试验利用固相萃取结合高效液相色谱法测定陕北狗头枣提取物中主要多酚成分的含量,同时基于1,1-二苯基-2-苦基肼(1,1-diphenyl-2-picryl-hydrazyl, DPPH)自由基清除试验、2,2′-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐[2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), ABTS]阳离子自由基清除试验和羟基自由基清除试验系统评价陕北狗头枣可食用部分多酚的抗氧化活性。研究结果表明,乙醇体积分数对陕北狗头枣可食用部分总多酚的提取效果具有显著影响。提取液中总多酚含量随着提取液中乙醇体积分数的增大呈先增加后减少的趋势,当乙醇体积分数为70%时,提取物中总多酚含量最高。此外,陕北狗头枣多酚提取物具有良好的抗氧化活性,清除DPPH、ABTS+和羟基自由基的IC50值分别为(3.28±0.03)、(3.65±0.04) mg/mL和(3.96±0.05) mg/mL,且其抗氧化能力与总多酚含量极显著相关(p<0.01)。
Abstract:The effects of ethanol concentration on the extraction quality of total polyphenols from the edible parts (peel and flesh) of ′Goutou′ jujube fruits in northern Shaanxi were investigated. The content of major polyphenols in the extract of the fruits was determined by solid-phase extraction coupled with high-performance liquid chromatography. The antioxidant activity of the polyphenols was comprehensively evaluated through 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay, and hydroxyl radical assay. The results demonstrated that ethanol concentration significantly influenced the extraction of total polyphenols. The total polyphenol content in the extract initially increased and then decreased as the ethanol concentration rose, reaching its peak at 70% ethanol. Furthermore,the polyphenol extract exhibited notable antioxidant activity, with IC50?values of (3.28±0.03), (3.65±0.04),and (3.96±0.05) mg/mL for DPPH, ABTS+, and hydroxyl radical scavenging, respectively. A correlation (p<0.01) was observed between the antioxidant capacity and the total polyphenol content.
