榆黄菇粗多糖的提取工艺优化及抗氧化、抗衰老活性分析

中文摘要: 为提高榆黄菇粗多糖得率,研究其抗氧化、抗衰老活性,该研究采用响应面法优化榆黄菇粗多糖提取工艺,并构建多指标评估体系。通过检测DPPH自由基清除率、羟自由基清除率及超氧阴离子自由基清除率,系统评价榆黄菇粗多糖的体外抗氧化活性;以秀丽隐杆线虫为模式生物,通过应激实验、生殖力实验、运动实验探究榆黄菇粗多糖的抗衰老活性。结果表明:榆黄菇粗多糖的最佳提取参数为提取温度53 ℃、提取时间2.4 h、液料比30∶1 (mL/g),此条件下的粗多糖得率为(18.76±0.16)%;榆黄菇粗多糖对DPPH自由基和羟自由基清除率较高,对超氧阴离子自由基的最大清除率为11.92%;榆黄菇粗多糖可以增强线虫热抵抗力,缓解H2O2和胡桃醌引起的氧化应激损伤,促进线虫产卵,增强线虫运动能力,提高线虫的身体弯曲频率和头部摆动频率,具有良好的抗衰老活性。

Abstract:To improve the yield of crude polysaccharides from Pleurotus citrinopileatus and study its antioxidant and anti-aging activities, the response surface method was applied to optimize the extraction process of crude polysaccharides from P. citrinopileatus. A multi-index evaluation system was constructed. The in vitro antioxidant activity of polysaccharides from P. citrinopileatus was systematically evaluated by detecting DPPH radical scavenging rate, hydroxyl radical scavenging rate, and superoxide anion scavenging rate. With Caenorhabditis elegans as the model organism, the anti-aging activity of polysaccharides from P. citrinopileatus was explored through stress experiments, reproductive experiments, and exercise experiments. The results showed that the extraction parameters for crude polysaccharides from P. citrinopileatus were optimal at an extraction temperature of 53 ℃, extraction time of 2.4 h, and liquid-to-material ratio of 30∶1(mL/g). Under these conditions, the yield of predicted crude polysaccharide was(18.76±0.16)%. Crude polysaccharides from P. citrinopileatus had a high clearance rate of DPPH free radicals and hydroxyl radicals, respectively, and the maximum scavenging rate for superoxide anion radicals was 11.92%. The crude polysaccharides from P. citrinopileatus can enhance the heat resistance of nematodes and alleviate oxidative stress damage caused by H2O2 and juglone. It can also promote the nematodes to lay eggs, enhance the motility, and increase the body bending frequency and head swing frequency, presenting good anti-aging activity.
