灰枣多糖提取工艺优化及其免疫活性

中文摘要: 为实现新疆灰枣资源的高值化应用,对灰枣多糖提取工艺及免疫活性进行研究。首先以多糖提取率为指标,比较热水、超声波辅助、微波辅助和低共熔体系4种方法的提取效果,并采用响应面法对提取方法进行优化;再经乙醇沉淀得到粗多糖,对粗多糖进行分离纯化,比较分离前后的免疫活性差异。结果显示,热水提取法的多糖提取率最高,为(3.93±0.25)%。最佳工艺条件为提取温度82 ℃、提取时间108 min、液料比17∶1(mL/g),此条件下,多糖提取率为(4.18±0.53)%。活性结果显示,粗多糖和纯化后多糖均有良好的活性,表明优化后的方法不仅能提高多糖提取率,同时获得了具有较强免疫活性的多糖。
Abstract:To realize the high-value application of Ziziphus jujuba cv. Huizao resources from Xinjiang, the extraction process and activity of polysaccharides from Ziziphus jujuba cv. Huizao were studied. Firstly, the polysaccharide extraction rate was set as an index to compare four extraction methods: hot water, ultrasound-assisted method, microwave-assisted method, and deep eutectic system. Next, the response surface method was employed to optimize the conditions of the optimal extraction method. Then, the alcohol precipitation was conducted to obtain the crude polysaccharide, which was isolated and purified. Finally, the immune activities were compared before and after the isolation. The results showed that the hot water extraction method had the highest polysaccharide extraction rate of (3.93±0.25)%. The optimal process conditions were an extraction temperature of 82 ℃, an extraction time of 108 min, and a liquid-material ratio of 17∶1 (mL/g). Under these conditions, the polysaccharide extraction rate was (4.18±0.53)%. The activity results showed that both crude polysaccharides and purified polysaccharides exhibited ideal activity, indicating that the optimized method improved the extraction rate while yielding polysaccharides with strong immune activity.
