花椒叶黄酮超声波辅助提取、纯化及其抗菌活性

中文摘要: 该研究旨在探索超声波辅助青花椒叶黄酮类物质提取工艺,并评估其抗菌活性。采用单因素试验结合正交试验,探究提取温度、提取时间、乙醇浓度、料液比、超声波功率以及脉冲超声间歇比对黄酮提取率的影响,采用大孔树脂对粗黄酮进行纯化,并采用牛津杯法研究花椒叶黄酮对大肠杆菌和金黄色葡萄球菌的抑菌效果。研究结果表明,超声波辅助提取花椒叶黄酮的最优工艺条件为提取温度40 ℃、提取时间25 min、乙醇浓度70%、料液比1∶30( g/mL)、超声波功率300 W和脉冲超声间歇比9∶3,在此条件下,黄酮提取率为14.93%,与对照相比,提取率提高了42.9%;大孔树脂吸附和解吸结果表明,AB-8号大孔树脂对花椒叶黄酮的纯化较高;纯化后的花椒叶黄酮对金黄色葡萄球菌有明显抑菌活性,最小抑菌浓度为0.4 mg/mL,但对大肠杆菌无明显抑制作用。

Abstract:This study aims to optimize the ultrasonic-assisted extraction process of flavonoids from Zanthoxylum bungeanum leaves and evaluate their antibacterial activities. Single-factor tests were combined with an orthogonal design to study the effects of extraction temperature, extraction time, ethanol concentration, solid-toliquid ratio, ultrasonic power, and ultrasonic interval ratio on the yield of flavonoids. The extracted flavonoids were purified by macroporous resins, and the inhibitory activities of the flavonoids on Escherichia coli and Staphylococcus aureus were measured by the Oxford cup method. The results showed that the optimal conditions for ultrasonic-assisted extraction of flavonoids from Z. bungeanum leaves were extraction temperature of 40 ℃, extraction time of 25 min, ethanol concentration of 70%, solid-to-liquid ratio of 1∶30 (g/mL), ultrasonic power of 300 W, and ultrasonic interval ratio of 9∶3, under which the yield of flavonoids was 14.93%,which was 42.9% higher than that of the control. The adsorption and desorption results of macroporous resins indicated that the AB-8 resin had a higher purification effect on Z. bungeanum leaf flavonoids. The purified flavonoids showed significant inhibitory activity against S. aureus, with a minimum inhibitory concentration of 0.4 mg/mL, while they demonstrated no significant inhibitory effect on E. coli.
