中文摘要: 以花生全植株为原料,对其各个部位的反式白藜芦醇提取方法进行研究,使用高效液相色谱法对其测定,并用液相色谱-质谱法进一步确认,同时考察方法的线性关系、重复性、精密度、稳定性和加标回收率等指标。结果显示,用80%甲醇溶液作提取剂,采用超声波加热辅助提取,花生中反式白藜芦醇提取完全,且在花生仁的提取过程中加入沉淀剂,目标组分与其他组分分离效果良好,反式白藜芦醇的测定不受杂质峰的干扰;反式白藜芦醇浓度在0.008 270~16.540 2 μg/mL时,线性回归方程为Y=79 086.176 7X+47.050 3(R2=1.000 0)线性关系良好;平均回收率为83.9%~104.0%,相对标准偏差为0.20%~1.94%。此方法准确可靠、重现性好,适合花生植株样品中反式白藜芦醇的测定。
Abstract:By taking the whole peanut plant as the raw material,the extraction method of trans-resveratrol from various parts of the plant was studied. The trans-resveratrol content was determined by high performance liquid chromatography(HPLC)and further confirmed by liquid chromatography-mass spectrometry(LC-MS).The linear relationship,repeatability,precision,stability,and spiked recovery rate of the method were investigated.The results showed that when 80% methanol solution was used,and ultrasonic heating-assisted extraction was adopted,the trans-resveratrol in various parts of the peanut plants was completely extracted. When the peanut kernels were extracted,and the precipitating agent was used,the target substance could be well separated from other components. The determination of trans-resveratrol was not disturbed by impurity peaks,In the concentration range of 0.008 270-16.540 2 μg/mL for trans-resveratrol,the linear regression equation was Y=79 086.176 7X+47.050 3,with a correlation coefficient R2=1.000 0,and the linear relationship was good. The average recovery rate was 83.9%-104.0%,and the relative standard deviation was 0.20%-1.94%.The method was accurate and reliable,and it had good reproducibility,which was suitable for the determination of trans-resveratrol in peanut plants.
