中文摘要: 该文以淫羊藿为原料,总黄酮得率为评价指标,利用超声波法辅助提取淫羊藿总黄酮,运用单因素试验和正交试验,优化淫羊藿总黄酮的提取工艺条件;通过DPPH自由基清除法、ABTS+自由基清除法、O2-自由基清除法和总还原能力试验,评价淫羊藿总黄酮的体外抗氧化活性。结果表明,淫羊藿总黄酮的最佳提取工艺为料液比1∶30(g/mL)、超声温度50 ℃、超声时间40 min、乙醇浓度60%。在此条件下,淫羊藿总黄酮得率为(10.39±0.01)%。抗氧化试验结果表明,淫羊藿总黄酮具有较强的总还原能力,当其浓度为1.040 mg/mL时,对DPPH自由基的清除率高达(75.15±0.43)%,对ABTS+自由基的清除率高达(71.19±1.38)%,对O2-自由基的清除率高达(81.82±0.95)%,表明淫羊藿总黄酮具有良好的抗氧化活性。
Abstract:This study utilized Epimedium as the raw material,with the yield of total flavonoids serving as the evaluation index. The ultrasonic method was employed to extract the total flavonoids from Epimedium. The extraction conditions were optimized through single-factor experiments and orthogonal experiments. The in vitro antioxidant activity of the total flavonoids was assessed using the DPPH free radical scavenging assay,ABTS+free radical scavenging assay,O2- free radical scavenging assay,and total reducing capacity test. The results demonstrated that the optimal extraction process for the total flavonoids from Epimedium involved a solid-to-liquid ratio of 1∶30(g/mL),an ultrasonic temperature of 50 ℃,an ultrasonic time of 40 min,and an ethanol concentration of 60%. Under these conditions,the yield of total flavonoids reached(10.39±0.01)%.The antioxidant results demonstrated that the total flavonoids extracted from Epimedium exhibited a robust total reducing capacity.Specifically,at a concentration of 1.040 mg/mL,the scavenging efficiency of DPPH free radicals reached(75.15±0.43)%;the scavenging efficiency of ABTS+free radicals reached(71.19±1.38)%,and the scavenging efficiency of O2-free radicals reached(81.82±0.95)%.These findings indicated that the total flavonoids from Epimedium possessed significant antioxidant activity.
