中文摘要: 为提高湿酱油渣的营养价值、改善其风味、拓宽湿酱油渣的利用途径,该文采用酶解法,以水解度为指标,考察加酶量、酶解温度、酶解pH 值、酶解时间和料液比对湿酱油渣酶解效果的影响。基于单因素和响应面试验,确定湿酱油渣最佳酶解条件为木瓜蛋白酶与菠萝蛋白酶复配比1∶2(体积比)、料液比1∶3 (g/mL)、加酶量340 U/g、酶解pH 值为7、酶解温度47 ℃、酶解时间4 h,在此条件下水解度达27.69%。酶解后的湿酱油渣口味与气味评分显著提高;蛋白质分子量发生明显改变,分子量小于244 Da 的占比达57.29%;总游离氨基酸含量由167.99 mg/100 g 升高至202.30 mg/100 g,其中7 种游离氨基酸含量显著升高。
Abstract:To improve the nutritional value,flavor,and utilization of wet soy sauce residue,the enzymatic hydrolysis method was used to investigate the effects of enzyme addition amount,enzymatic hydrolysis temperature,enzymatic hydrolysis pH,enzymatic hydrolysis time and solid-liquid ratio on the enzymatic hydrolysis result of wet soy sauce residue by enzymatic hydrolysis method,with hydrolysis degree as an index. Based on the single factor and response surface experiment,the optimal enzymatic hydrolysis conditions of wet soy sauce residue were determined to be as follows:the papain to bromelain ratio of 1∶2 (volume ratio),the solid-liquid ratio of 1∶3 (g/mL),the enzyme addition amount of 340 U/g,the enzymatic hydrolysis pH value of 7,the enzymatic hydrolysis temperature of 47 ℃,and the enzymatic hydrolysis time of 4 h,and the hydrolysis degree of 27.69%. After enzymatic hydrolysis,wet soy sauce residue′s taste and odor scores were significantly improved.The molecular weight of the protein was obviously changed,with the molecular weight less than 244 Da accounting for 57.29%. The total content of free amino acids was increased from 167.99 mg/100 g to 202.30 mg/100 g,with the contents of 7 free amino acids increasing significantly.
