中文摘要: 为研究番荔枝果肉中膳食纤维的提取方法,并明确其结构和理化特性,该文采用超声波辅助酶法提取番荔枝果肉可溶性膳食纤维(Annona squamosa pulp soluble dietary fiber,AP-SDF),对超声时间、热稳定α-淀粉酶添加量、碱性蛋白酶添加量和水浴时间对AP-SDF 得率的影响进行考察,通过单因素和正交试验优化提取工艺条件,并通过X-射线衍射技术、傅里叶变换红外光谱技术等对AP-SDF 的结构和理化特性进行分析。结果表明:AP-SDF 的最佳提取条件为超声时间30 min、热稳定α-淀粉酶添加量3%、碱性蛋白酶添加量1.0%、水浴时间60 min,此条件下APSDF 得率为(15.78±0.37)%。AP-SDF 含有羧基、羟基、醛基等活性功能基团,且结晶度较低,质地松软,有利于增加水合性能,表现出较强的持水力、持油力和膨胀力,是优质的膳食纤维,具有较大的开发价值。
Abstract:To established extraction methods and exploring the structure and physicochemical properties of dietary fiber from Annona squamosa Pulp,the ultrasound-assisted enzymatic method was used to extract soluble dietary fiber from Annona squamosa Pulp (AP-SDF). The factors influencing the yield of AP-SDF,such as ultrasound time,dosage of thermostable α-amylase,dosage of alkaline protease,and water bath time were investigated,and the process conditions were optimized by single-factor test and orthogonal test. In addition,X-ray diffraction,Fourier transform infrared spectrum,and other technologies were employed to reveal the structure and physicochemical properties of AP-SDF. The result showed that the optimized extraction conditions for AP-SDF were ultrasound time of 30 min,3% dosage of thermostable α-amylase,1.0% dosage of alkaline protease,and water bath time of 60 min. Under these conditions,the yield of AP-SDF was (15.78±0.37)%. AP-SDF contained active functional groups such as carboxyl,hydroxyl,and aldehyde groups. It had a low crystallinity and a soft texture,which was conducive to improving hydration performance. It showed strong water-holding capacity,oil-holding capacity,and expansion capacity,serving as a high-quality dietary fiber and exhibiting great development value.
