中文摘要: 为探究海参酶解物对急性酒精致胃黏膜损伤的缓解作用,该文采用傅里叶变换红外光谱、高效凝胶渗透色谱以及液相色谱-质谱联用等技术对海参酶解物的官能团种类、分子量大小以及海参多糖含量等指标进行测定及分析,再通过高压均质工艺将海参酶解物混溶于白酒中,构建胃黏膜损伤大鼠模型。结果表明:海参酶解物中海参多糖含量为8.53%,蛋白质分子量分布在2.5 kDa 和1.0 kDa 附近的肽段,另外还有100.0 Da 左右的游离氨基酸,总氨基酸含量为89.96%,蛋白质含量为77.50%。大鼠胃组织切片的病理学结果以及氧化应激指标的结果表明,高剂量海参酒(1.0 g/100 mL)能够缓解酒精引起的胃黏膜损伤。
Abstract:The effect of a sea cucumber enzyme hydrolysate on acute alcoholic gastric mucosa injury was studied. A sea cucumber enzyme hydrolysate with high polysaccharide content was prepared for the production of sea cucumber liquor. Fourier transform infrared spectroscopy,high performance gel permeation chromatography,and liquid chromatography-mass spectrometry were utilized to determine and analyze the functional groups,molecular weights,and polysaccharide content of the sea cucumber hydrolysate. Subsequently,the high-pressure homogenization process was employed to mix the sea cucumber enzyme hydrolysate into the liquor.Then,a rat model of gastric mucosal injury was established to explore the effect of adding the sea cucumber enzyme hydrolysate to the liquor on alcoholic gastric mucosal injury. The results demonstrated that the polysaccharide content of the sea cucumber enzyme hydrolysate was 8.53%. The peptides in the hydrolysate had the molecular weights of about 2.5 kDa and 1.0 kDa,and there was a small amount of free amino acids of approximately 100.0 Da. The total amino acid content was 89.96% and the protein content was 77.50%. The pathological results and oxidative stress indicators of the rat gastric tissue sections indicated that adding highdose sea cucumber enzyme hydrolysate(1.0 g/100 mL)into liquor alleviated the gastric mucosal injury induced by alcohol.
