中文摘要: 为探寻迷迭香精油不同蒸馏段的化学成分特征与抑菌活性,该研究以迷迭香鲜条为原料,采用水蒸气蒸馏法制备得到7 个不同蒸馏段精油,利用气相色谱-质谱(gas chromatography-mass spectrometer,GC-MS)法对各精油的成分及相对含量进行测定,采用滤纸片法测定各精油对大肠杆菌、金黄色葡萄球菌的抑菌活性。试验结果显示,随着蒸馏的进行,精油得率显著递减。7 个蒸馏段共鉴定出52 种成分,主要为萜烯、酮、环氧化合物、萜烯醇、酚、醛及酯类化合物。其中,36 个化合物为不同段精油的共有成分,反式桧烯水合物与3-羟基-β-环柠檬醛随着蒸馏的进行,其相对含量逐渐从0.09%~0.15%减少到0%,10 种化合物随着蒸馏的进行,相对含量从0%增加至0.2%~2.4%。对于大肠杆菌和金黄色葡萄球菌,7 个蒸馏段精油均具有抑菌活性,随着蒸馏的进行,精油抑菌活性逐渐增加,其中第七蒸馏段表现出最强的抑菌活性。综上,迷迭香不同时间蒸馏段精油出油量、化学成分及抑菌活性差异明显。
Abstract:To investigate the chemical composition characteristics and antimicrobial activities of different rosemary essential oils from different distillation stages,fresh rosemary stems were taken as raw material in this study,and steam distillation was employed to obtain essential oils from seven distinct distillation stages. The components and their relative contents in essential oils from each distillation stage were determined by gas chromatography-mass spectrometry(GC-MS).Their antimicrobial activities against Escherichia coli and Staphylococcus aureus were evaluated through filter paper method.The findings indicated a significant decrease in essential oil yield as distillation progressed. 52 components were identified among the seven distillation stages,mainly comprised of terpenes,ketones,epoxy compounds,terpenols,phenols,aldehydes,and ester compounds. Of these,36 compounds were common constituents across different distillation periods. The relative contents of trans-sabinene hydrate and 3-hydroxy-β-cyclocitral gradually decreased from 0.09%-0.15% to 0%as the distillation proceeded,while the relative contents of 10 compounds increased from 0% to 0.2%-2.4%.Essential oils from all seven distillation stages exhibited antimicrobial activity against both E. coli and S. aureus. The antimicrobial activity of the essential oils increased gradually as the distillation progressed,with the strongest antimicrobial activity occurring at the seventh distillation stage. In summary,the essential oils obtained from different distillation stages of rosemary demonstrated significant variations in oil yield,chemical composition,and antimicrobial activity.
