HS-SPME 萃取煳辣椒挥发性物质的条件优化及其成分解析

中文摘要: 为解析煳辣椒挥发性物质,该研究选取贵州煳辣椒为研究对象,以总峰面积为指标,采用顶空固相微萃取法(head-space solid phase microextraction,HS-SPME)对其挥发性物质进行萃取并优化萃取条件,建立适用于煳辣椒挥发性成分分析的气相色谱-质谱联用方法。结果表明,贵州煳辣椒的香气属性以香料、咖啡和烟熏味为主,HS-SPME萃取煳辣椒挥发性物质的最佳条件为萃取温度70 ℃、平衡时间10 min、饱和NaCl 溶液5 mL、萃取时间50 min。在此最佳的萃取条件下,共检出11 类94 种化合物,主要包含萜烯类(27.66%)、醛类(18.09%)、含硫化合物(10.64%)、含氮化合物(9.57%)、酮类(8.51%)、醇类(7.45%)、酯类(7.45%)、呋喃类(3.19%)、酚类(3.19%)、酸类(2.13%)以及烃类化合物(2.13%),半定量结果表明主要的挥发性物质为3-糠醛、反式-2,4-葵二烯醛、苯甲醛、2-乙酰基吡咯、2-甲基吡嗪、N-甲基吡咯、5-甲基-2-乙酰基呋喃、乙酸芳樟酯、月桂烯、芳樟醇等化合物。该研究为煳辣椒风味解析及其后期品质提升提供理论依据。
Abstract:The volatile compounds of Hu pepper cultivated in Guizhou were studied. With the total peak area as an indicator,the headspace solid-phase microextraction (HS-SPME) conditions were optimized,and a method based on gas chromatography-mass spectrometry suitable for the analysis of volatile compounds in Hu pepper was established. The results showed that Hu pepper in Guizhou mainly presented spice,coffee,and smoky flavors. The optimal conditions for extracting volatile compounds from Hu pepper were extraction temperature of 70 ℃,incubation time of 10 min,saturated NaCl solution of 5 mL,and extraction time of 50 min.Under the optimal extraction conditions,a total of 94 compounds belonging to 11 categories were detected,including terpenes (27.66%),aldehydes (18.09%),sulfur - containing compounds (10.64%),nitrogen -containing compounds (9.57%),ketones (8.51%),alcohols (7.45%),esters (7.45%),furans (3.19%),phenols (3.19%),acids (2.13%),and hydrocarbons (2.13%). The main volatile compounds were 3-furfural,trans,trans-2,4 decadienal,benzaldehyde,2-acetylpyrrole,2-methylpyrazine,N-methylpyrrole,5-methyl-2-acetylfuran,linalool acetate,myrcene,and linalool. The established extraction method in this study provided a theoretical basis for analyzing the flavor and improving the quality of Hu pepper.
