中文摘要: 为建立辣椒生物碱与辣椒色素分离纯化工艺,并探讨辣椒生物碱的杀虫及保鲜作用,该文通过考察大孔吸附树脂的吸附与解吸作用,辣椒生物碱和辣椒色素分别使用不同的树脂进行纯化,并优化其工艺参数;以杀虫和水果保鲜试验,开展辣椒生物碱的杀虫和水果保鲜作用研究。结果表明,先用ME-1 树脂分离纯化辣椒生物碱后再用聚酰胺树脂纯化辣椒红色素可以同时获得辣椒中2 种主要活性成分;纯化后的辣椒生物碱含量为82.796 mg/g,辣椒色素色价为603,分别是辣椒粗提取物的61 倍和7 倍;杀虫试验表明,辣椒碱、二氢辣椒碱杀虫作用与阳性药物相当,当二者浓度为1.0 mg/mL 时杀虫率可达100%;辣椒生物碱纯化产物为2.0 mg/mL 时,杀虫率可达80%以上;水果保鲜试验表明,辣椒碱、二氢辣椒碱为2.0 mg/mL、辣椒生物碱纯化产物为6.0 mg/mL 时,水果出现腐烂的时间延长了3~7 d,失重率、可溶性固形物及维生素C 含量变化小于空白组,且与4 ℃储藏水果的变化幅度相近。综上,通过2 种树脂能同时分离纯化辣椒中辣椒生物碱和辣椒色素,辣椒生物碱具有良好的杀虫和水果保鲜作用。
Abstract:To establish a separation and purification process for capsicum alkaloids and capsicum pigments and explore the insecticidal and preservative effects of capsicum alkaloids,this article used different resins to purify capsicum alkaloids and capsicum pigments,respectively,after investigating the adsorption and desorption effects of macroporous adsorption resins,and optimized the process parameters. Insecticidal and fruit preservation experiments were conducted to study the effects of capsicum alkaloids. The results indicated that the sequential use of ME-1 resin for the separation and purification of capsicum alkaloids followed by polyamide resin for the purification of capsicum pigments allowed for the simultaneous acquisition of two major active components from capsicum. The purified capsicum alkaloid content reached 82.796 mg/g,while the color value of the capsicum pigment was 603,representing a 61-fold and 7-fold increase compared to the crude capsicum extract,respectively. The insecticidal tests indicated that the insecticidal effects of capsaicin and dihydrocapsaicin were comparable to those of positive drugs. When the concentration of these two compounds reached 1.0 mg/mL,the insecticidal rate reached 100%;and when the concentration of the purified capsicum alkaloids reached 2.0 mg/mL,the insecticidal rate can reach over 80%. In the fruit preservation tests,when capsaicin and dihydrocapsaicin were used at a concentration of 2.0 mg/mL and the purified capsicum alkaloid product was at 6.0 mg/mL,the shelf life of the fruit was extended by 3 d to 7 d. The changes in weight loss rate,soluble solids,and VC content were lower than those observed in the control group and were similar to the changes in fruit stored at 4 ℃. In summary,capsicum alkaloids and capsicum pigments can be separated and purified simultaneously by two types of resins,and capsicum alkaloids demonstrate excellent insecticidal and fruit preservation effects.
