沙棘籽粕制备抑菌肽的酶解工艺及稳定性

中文摘要: 通过比较8 种沙棘籽粕蛋白酶解物对金黄色葡萄球菌、枯草芽孢杆菌、沙门氏菌、大肠杆菌的抑菌活性,并以抑菌圈直径为评价指标,筛选出胰蛋白酶作为制备抑菌肽受试蛋白酶。利用单因素结合响应面试验,确定沙棘籽粕抑菌肽最佳制备条件为酶解pH8.90、酶解温度39.5 ℃、加酶量6 240 U/g、酶解时间2.2 h。在此条件下,沙棘籽粕抑菌肽的抑菌圈直径模型预测值为22.60 mm,实测值为22.33 mm。通过对其温度、酸碱、紫外照射、反复冻融处理后抑菌活性的测试,发现沙棘籽粕抑菌肽具有较好的抑菌稳定性。
Abstract:The antibacterial activities of zymolytes from eight types of Hippophae rhamnoides seed meal were compared against Staphylococcus aureus,Bacillus subtilis,Salmonella,and Escherichia coli. With the diameter of inhibition as the evaluation criterion,trypsin was selected as the enzyme for the preparation of antibacterial peptides. To ascertain the optimal conditions for the preparation of antibacterial peptides,both single-factor and response surface experiments were conducted. The identified optimal conditions were a pH of 8.90,a temperature of 39.5 ℃,an enzyme addition of 6 240 U/g,and a reaction time of 2.2 h. Under these conditions,the diameter of inhibition for the antibacterial peptides of Hippophae rhamnoides seed meal was estimated at 22.60 mm,with an actual measured diameter of 22.33 mm. Further testing of the antibacterial activity of these peptides under variable conditions such as temperature fluctuations,pH changes,ultraviolet radiation exposure,and repeated freeze-thaw cycles demonstrated that antibacterial peptides of Hippophae rhamnoides seed meal exhibited substantial antibacterial stability.
