中文摘要: 以发酵万寿菊渣为原料,采用超声辅助法提取万寿菊总黄酮粗提物,利用响应面试验对其提取工艺进行优化。通过离心水洗法对粗提物进行纯化,并测定其纯度。通过测定DPPH 自由基和ABTS+自由基的清除率对万寿菊总黄酮提取物进行抗氧化活性测定。结果表明,万寿菊总黄酮的最佳提取工艺为料液比1∶22 (g/mL)、超声温度48 ℃、乙醇体积分数71%、超声时间63 min,在该工艺条件下万寿菊总黄酮的提取率为6.69%。经纯化后,纯度最高可达到60.63%。体外抗氧化结果表明:万寿菊总黄酮具有良好的DPPH 自由基和ABTS+自由基清除能力,当纯化万寿菊总黄酮质量浓度为50 μg/mL 时,对DPPH 自由基和ABTS+自由基的清除能力均趋近于相同浓度下的维生素C。
Abstract:Total flavonoids were extracted from fermented marigold residue by ultrasound-assisted extraction method,and the extraction process was optimized by the response surface methodology. The crude extract was purified by centrifugation and water washing and its purity was determined. The antioxidant activity of total flavonoids from marigold residue was measured based on the scavenging rates of DPPH and ABTS+ free radicals.The results showed that the optimum extraction process of total flavonoids from marigold residue was as follows:solid-to-liquid ratio of 1∶22 (g/mL),ultrasonic temperature of 48 ℃,ethanol volume fraction of 71%,and ultrasonic time of 63 min. Under this process conditions,the extraction rate of total flavonoids from marigold residue was 6.69%. After purification,the maximum purity could reach to 60.63%. The extracted total flavonoids had strong scavenging activities against DPPH and ABTS+ free radicals. At the mass concentration of 50 μg/mL,the purified total flavonoids from marigold residue showed the DPPH and ABTS+ free radical scavenging activities close to that of vitamin C at the same concentration.
