中文摘要: 为探究小麦粉面团醒发过程中淀粉颗粒和面团结构的变化,以市售小麦粉为原料,面粉与水按料液比2∶1 (g/mL)配比,利用pH 仪、紫外分光光度计和显微镜等仪器检测醒发过程中(5、10、15、20、25 min)面团的pH值、淀粉颗粒、可提取蛋白质含量、干湿面筋含量以及面团微观结构等指标。结果表明,面团在醒发过程中,面团整体呈酸性,pH 值先降低后升高;淀粉颗粒直径逐渐增加,在20 min 时直径达到最大;面团中水溶性蛋白含量呈现高低波动,而盐溶性蛋白含量呈持续增加趋势,醇溶蛋白含量呈先降低后升高再降低的趋势,十二烷基硫酸钠(sodium dodecyl sulfate,SDS)可溶性麦谷蛋白含量呈现出逐渐升高的趋势;淀粉颗粒直径与pH 值、透光率和干面筋含量之间呈显著相关,相关系数分别为-0.914、-0.935 和0.910(p<0.05)。面团醒发时间为15~25 min 时,淀粉颗粒变化最为明显,面筋网络最易形成且稳定,是小麦粉面团发酵的较佳时间段。
Abstract:To explore the changes in starch particles and structure of wheat flour dough during fermentation,commercially available wheat flour was used as raw material,and the ratio of flour and water was 2∶1 (g/mL).The pH meter,ultraviolet spectrophotometer,and microscope were used to detect the pH,starch particles,extractable protein content,dry and wet gluten content,and microstructure of the dough during fermentation (5,10,15,20,and 25 min). The results showed that the dough became acidic during fermentation,and the pH value of the dough decreased first and then increased. The diameter of starch particles was gradually increased and reached the maximum at 20 min. The amount of water-soluble protein in the dough fluctuated,while the salt-soluble protein increased continuously. The gliadin content became low at first and then high,following another decrease. The SDS-soluble glutenin showed a trend from low content to high content. The diameter of starch particles was significantly correlated with pH value,light transmittance,and dry gluten content,and the correlation coefficients were-0.914,-0.935,and 0.910,respectively (p<0.05). With in 15-25 min of dough fermentation,the change in starch particles was the most obvious,and the gluten network was the easiest to form and stable,indicating the best time for wheat flour dough fermentation.
