中文摘要: 原儿茶酸(protocatechuic acid,PCA)是一种分布广泛的天然酚类化合物,也是花色苷、原花色苷等复杂多酚的主要代谢物之一,具有抗氧化、神经保护等功能。采用多光谱技术研究PCA 与卵白蛋白(ovalbumin,OVA)的相互作用及其对ABTS+自由基清除能力的影响。结果显示,PCA 对OVA 本征荧光产生猝灭作用;猝灭常数Ksv 随温度升高而降低,表明相互作用为静态猝灭机制;PCA 与OVA 存在一个结合位点,结合常数约为104 量级;结合属于自发产生的吸热反应,疏水作用是主导作用力;相互作用后Tyr 残基周围微环境极性增加、疏水性降低,Trp 残基所处微环境没有变化。PCA-OVA 复合物ABTS+自由基清除能力较PCA 和OVA 有显著提高(p<0.05)。因此,PCA 与OVA 的相互作用使OVA 构象发生改变,对ABTS+自由基清除能力具有一定影响。
Abstract:Protocatechuic acid(PCA)is a natural phenolic compound with wide distribution.It is also a main metabolite of complex polyphenols including anthocyanin and protocatechuic acid,which has the antioxidant and neuroprotective functions.Multispectral imaging was used to measure the interaction between PCA and ovalbumin(OVA)and its effect on the antioxidant properties of PCA against ABTS+ radical.The results showed as follows.The emission intensity of OVA was quenched by PCA,and the quenching constant(Ksv)decreased with the increasing temperature,indicating the interaction between PCA and OVA was carried out by a static quenching mechanism.One binding site existed between PCA and OVA,with binding constants of about 104.The bonding was a spontaneously generated heat absorption reaction with hydrophobicity as the dominant force.The microenvironment around the Tyr residue increased in polarity and decreased in hydrophobicity after the interaction,while the microenvironment in which the Trp residue was located did not change.The ABTS+ radical scavenging ability of PCA-OVA complex wassignificantly improved compared with that of PCA and OVA(P<0.05).Therefore,the interaction between PCA and OVA changes the conformation of OVA,and also has certain effect in scavenging ABTS+ radical.
