中文摘要: 该文对大西洋雪松精油的组分、体外抗菌及抗炎活性进行研究。利用气相色谱-质谱联用技术(gas chromatograph-mass spectrometer,GC-MS)对其进行分析,采用峰面积归一化法计算相对含量;使用牛津杯法测定其对金黄色葡萄球菌、大肠埃希氏菌及白色念珠菌的抑菌效果;建立脂多糖(lipopolysaccharide,LPS)诱导的RAW264.7 炎症模型,测定细胞上清液中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的含量。结果表明,GC-MS 共检出30 个峰,鉴定出18 种化合物,占总峰面积的87.58%,萜烯类化合物占83.26%,其中含量较高的物质为(+)-香橙烯(31.56%)、长叶环烯(20.29%)、(-)-α-古芸烯(12.90%);大西洋雪松精油对金黄色葡萄球菌、大肠埃希氏菌、白色念珠菌均有一定的抑菌作用且该精油能在安全浓度下有效抑制RAW264.7 细胞iNOS mRNA 的表达,且对TNF-α mRNA 的表达及上清液中TNF-α 的分泌影响显著。大西洋雪松精油具有一定的抗菌和抑制氧化应激炎症相关酶iNOS 的活力,GCMS 可作为其快速、便捷的质量分析方法。

Abstract:To study the components and in vitro antibacterial and anti-inflammatory activities of essential oils from Cedrus atlantica.Gas chromatography-mass spectrometry(GC-MS)was performed to analyze the essential oils,and peak area normalization method was used to calculate the relative content. Oxford cup method was employed to determine the antibacterial effect of the essential oils on Staphylococcus aureus,Escherichia coli and Candida albicans. Then an LPS-induced RAW264.7 inflammation model was established to determine the content of tumor necrosis factor-α(TNF-α)in the cell supernatant.Thirty peaks were detected by GC-MS,and 18 compounds were identified,accounting for 87.58% of the total peak area.Among them,terpenes accounted for 83.26%,of which(+)-aromadendrene(31.56%),longicyclene(20.29%)and(-)-α-gurjunene(12.90%)had higher content. C. atlantica essential oils had certain antibacterial effects on S. aureus,E. coli,and C.albicans.Moreover,the expression of iNOS mRNA in RAW264.7 cells was markedly inhibited at a safe concentration of the essential oils,and the mRNA expression of TNF-α and the secretion of TNF-α in supernatant were significantly affected. C. atlantica essential oils had certain antibacterial activities and can inhibit oxidative stress and inflammation-related enzyme iNOS,and GC-MS was a fast and convenient method for quality analysis of the essential oils.
