中文摘要: 对刺梨醇提物的降脂活性及其作用机制进行研究。取健康成年昆明种小鼠48只,随机分为正常组、模型组、刺梨醇提物高、中、低剂量组、辛伐他汀组。模型组给予高脂饲料饲养30 d,造成食源性肥胖模型。造模同时灌胃给予相应药物,连续给药30 d。自造模20 d起,与正常组小鼠相比,模型组小鼠体质量和饮食量显著上升(P<0.05);与模型组相比,刺梨醇提物高、中剂量组小鼠体质量、饮食量均显著降低(P<0.05);造模30 d辛伐他汀组小鼠体质量与模型组相比显著降低(P<0.05),饮食量变化无显著性差异。与模型组相比,刺梨醇提物高、中剂量组、辛伐他汀组小鼠血清总胆固醇、甘油三酯、低密度脂蛋白胆固醇均显著降低(P<0.05);油红O染色显示刺梨醇提物各剂量组、辛伐他汀组肝脏脂肪含量显著降低;血清瘦素酶联免疫吸附测定结果显示,模型组小鼠血清瘦素较正常组显著升高(P<0.05),刺梨醇提物高、中剂量组小鼠血清瘦素较模型组相比显著降低(P<0.05)。刺梨醇提物可显著降低食源性肥胖小鼠体质量、饮食量和血脂含量,其作用机制与降低食源性肥胖小鼠血清瘦素有关。
Abstract:This article explored the lipid-lowering activity of Rosa roxburghii Tratt.Ethanol extracts and the mechanisms.After extracting Rosa roxburghii Tratt.,48 healthy adult Kunming(KM)mice were randomly divided into a normal group,a model group,high,medium,and low-dose R.roxburghii Tratt.ethanol extract groups,and a simvastatin group.The model group was given a high-fat diet for 30 d to induce the mouse model of food-induced obesity,and at the same time,corresponding drugs were given by gavage for 30 d.After 20 d of modeling,the body mass and dietary intake of mice in the model group were increased significantly as compared with the normal group(P<0.05),and the body mass and dietary intake of mice in the high and medium-dose R.roxburghii Tratt.ethanol extract groups were significantly reduced as compared with the model group(P<0.05).The body mass of mice in the simvastatin group was significantly lower than that of the model group(P<0.05)after 30 d of modeling,and there was no significant difference in dietary intake.As compared with the model group,the serum total cholesterol(TC),triglyceride(TG),and low-density lipoprotein cholesterol(LDL-C)of mice in the high and medium-dose R.roxburghii Tratt.ethanol extract groups and the simvastatin groups were significantly reduced(P<0.05).The oil red O staining showed the hepatic fat content of mice in the R.roxburghii Tratt.ethanol extract groups and simvastatin group was significantly reduced.The results of the enzyme-linked immunosorbent assay(ELISA)showed that the serum leptin of mice in the model group was significantly higher than that of the normal group(P<0.05),and the serum leptin of mice in the high and medium-dose R.roxburghii Tratt.ethanol extract groups was significantly lower than that of the model group(P<0.05).In conclusion,R.roxburghii Tratt.ethanol extracts can significantly reduce the body mass,dietary intake,and blood lipid content of food-induced obese mice,and its mechanism is related to the reduction of serum leptin in food-induced obese mice.
