中文摘要: 为探究碱性条件(pH9.0)下反式肉桂酸(trans-cinnamic acid,TCA)对β-伴大豆球蛋白抗原性、抗原表位及蛋白结构的影响,利用间接酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法评估互作后蛋白与免疫球蛋白G(immunoglobulin G,IgG)及表位抗体的结合能力;通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)测定蛋白分子量变化;利用紫外、荧光、红外光谱表征蛋白的结构变化。结果表明:在碱性条件下TCA与β-伴大豆球蛋白之间存在相互作用,且TCA可以降低蛋白的抗原性。互作后蛋白质的二、三级结构有显著变化,并降低了蛋白的表面疏水性,随着TCA添加浓度的增加,表面疏水性变化更加明显。
Abstract:This study aimed to explore the effect of trans-cinnamic acid(TCA)on the antigenicity,epitope and protein structure of β-conglycinin under alkaline conditions(pH9.0).The indirect enzyme-linked immunosorbent assay(ELISA)was used to evaluate the binding ability of the interacted protein to immunoglobulin G(IgG)and epitope antibodies.The sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)was used to determine the molecular weight of the protein.The structural changes in the protein were characterized by ultraviolet,fluorescence and infrared spectroscopy.The results showed that there was an interaction between TCA and β -conglycinin under alkaline conditions,and TCA reduced the antigenicity of the protein.After interaction,the secondary and tertiary structures of the protein changed significantly,and the surface hydrophobicity of the protein was reduced.With the increase of the concentration of TCA,the surface hydrophobicity changed more obviously.
