中文摘要: 对提取纯化的葛根素进行生物活性研究,利用高效液相色谱(high performance liquid chromatography,HPLC)、红外光谱(infrared spectrum,IR)进行含量和结构测试,用清除自由基的方法评价纯化后葛根素样品体外抗氧化活性,用滤纸片法测定其体外抑菌活性,利用酶标仪测定葛根素样品最低抑菌浓度。结果表明:纯化后HPLC法测定葛根素含量为62.03%,一定浓度的葛根素对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐阳离子 [2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)cation,ABTS+]自由基、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基和羟基自由基清除率均达80%以上;6种菌体的抑菌效果为大肠杆菌>金色葡萄球菌>沙门氏菌>枯草芽孢杆菌,但对绳状青霉菌和黑曲霉菌没有抑菌活性,葛根素样品对金色葡萄球菌和大肠杆菌的最低抑菌浓度分别为8.5、4.5 mg/mL。表现出良好的体外抗氧化和抑菌生物活性。

Abstract:The extracted and purified puerarin was analyzed for biological activity,and the content and structure were determined using high-performance liquid chromatography(HPLC)and infrared spectroscopy(IR).The antioxidant activity in vitro of purified puerarin was evaluated by the scavenging method of free radicals.The antibacterial activity in vitro was determined by the filter paper method,and the minimum inhibitory concentration of puerarin was determined by the microplate reader.The results showed that after purification,the content of puerarin determined by HPLC was 62.03%,and the scavenging rates of puerarin to 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate) cation(ABTS+)free radicals,1,1-diphenyl-2-picrylhydrazyl(DPPH)free radicals,and hydroxyl radicals at a certain concentration were more than 80%.The antibacterial effect on six strains was in the sequence of Escherichia coli>Staphylococcus aureus>Salmonella>Bacillus subtilis,but no bacteriostatic activity against Penicillium funiculosum and Aspergillus niger was observed.The minimum inhibitory concentrations of puerarin against S.aureus and E.coli were 8.5 mg/mL and 4.50 mg/mL,respectively,showing good in vitro antioxidant and bacteriostatic biological activities.
