中文摘要: 抑制胰脂肪酶的活性能够有效地减少肥胖的发生。该研究从酶学动力学、荧光猝灭和分子对接的不同角度研究柚皮素对胰脂肪酶的抑制作用。抑制酶活性试验结果表明,柚皮素以可逆竞争性方式抑制胰脂肪酶的活性,且半数抑制浓度为2.413 mmol/L。荧光猝灭试验结果显示,柚皮素与胰脂肪酶结合后酶的荧光被猝灭,胰脂肪酶的构象发生变化。分子对接结果显示,柚皮素与胰脂肪酶形成了酶-抑制剂复合物,阻止了酶和底物的结合,从而导致酶催化活性降低。
Abstract:Inhibition of pancreatic lipase activity can effectively reduce the incidence of obesity.In this study,the inhibitory effect of naringenin on pancreatic lipase was studied through enzymatic kinetics,fluorescence quenching,and molecular docking.The enzyme activity experiment showed that naringenin inhibited pancreatic lipase activity in a reversible competitive manner,with the half inhibitory concentration of 2.413 mmol/L.The fluorescence quenching experiment showed that the binding with naringenin quenched the fluorescence and changed the conformation of pancreatic lipase.Molecular docking results showed that naringenin and pancreatic lipase formed an enzyme-inhibitor complex,which prevented the binding of enzyme with substrate,decreasing the enzyme catalytic activity.
