中文摘要: 黄曲霉毒素B1(aflatoxin B1,AFB1)是黄曲霉和寄生曲霉等真菌产生的有毒次级代谢物,具有很强的毒性以及致癌性和致畸性。因此构建操作简便、快速可靠的AFB1检测方法具有重要的研究意义。该文以核酸适配体作为分子探针,利用氧化石墨烯(graphene oxide,GO)猝灭荧光和吸附单链DNA的特点构建一种荧光生物传感器用来快速检测AFB1。研究结果表明,在最佳检测条件下,该方法能在30 min左右完成检测,在2.37 ng/mL~200.00 ng/mL呈现良好的对数关系(R2=0.972 7),其检测限为2.37 ng/mL,花生样品加标回收试验表明回收率为85.0% ~114.4% 。
Abstract:Aflatoxin B1(AFB1) is a toxic secondary metabolite produced by fungi such as Aspergillus flavus and Aspergillus parasiticus,which has strong toxicity,carcinogenicity and teratogenicity.Therefore,it is of great importance to develop a simple,fast and reliable detection assay for AFB1.In this study,a rapid fluorescence assay for AFB1detection was constructed based on graphene oxide(GO)nanosheet and aptamer probe.The results showed that under the optimal conditions,the assay could complete in about 30 min and exhibited a good logarithmic relationship(R2=0.972 7)in range of 2.37 ng/mL-200.00 ng/mL,with the detection limit of 2.37 ng/mL,and the recovery rate of peanut samples was 85.0% -114.4% .
