中文摘要: 为优化黄芪中活性成分的提取方法,采用4种单一提取法包括亚临界提取、超声辅助提取、酶解提取、水磨提取和3种联合提取包括超声辅助提取、酶解提取及水磨提取分别联合亚临界提取对黄芪中活性成分进行提取,并对比其抗氧化活性。结果表明:亚临界提取及联合提取对黄芪中总皂苷的提取效果较好,其中超声辅助、酶解辅助、水磨以及亚临界提取总皂苷得率分别为3.71% 、2.92% 、1.72% 和1.91% 。酶解辅助和水磨提取对总黄酮提取效果较好,其得率分别为1.04% 和2.20% 。酶解提取黄芪多糖的提取效果较好,多糖得率为6.05% 。不同提取方法对提取后黄芪残渣的组织结构均具有显著性影响。亚临界及联合提取所得提取液体外抗氧化能力均强于其它提取方法,其DPPH、ABTS+自由基清除能力及铁离子还原能力为其它提取方法的8倍~16倍、5倍~8倍、36倍~115倍,由相关性分析得出提取液体外抗氧化能力与总皂苷得率呈显著正相关性,与黄芪皂苷II含量以及美拉德反应水平呈极显著正相关性。
Abstract:To optimize the extraction method of active components from Astragalus radix,the active components of A.radix were extracted and the antioxidant activities were compared by four single extraction methods(subcritical water extraction,ultrasonic extraction,enzymatic extraction,and water grinding extraction) and three combining extraction methods (ultrasonic,enzymatic,and water grinding,combining subcritical water extraction,respectively).The results showed that subcritical water extraction and combining extraction methods were superior to other methods on the extraction of saponins.The total extraction yield of saponins by ultrasonic,enzymatic,water grinding,and subcritical water extractions was 3.71% ,2.92% ,1.72% ,and 1.91% ,respectively.Both the enzymatic and water grinding extractions had better effects on the extraction of total flavonoids,with the value of 1.04% and 2.20% ,respectively.The enzymatic extraction was the most effective in the extraction of polysaccharides,with the value of 6.05% .The effect of different extraction methods on the tissue structure of the residue of A.Radix after extraction was significant.The antioxidant capacity of A.Radix extract obtained by subcritical water extraction and combining extraction methods were stronger than other extraction methods.The DPPH and ABTS+radical scavenging abilities and the ferric reducing ability were 8 times-16 times,5 times-8 times,and 36 times-115 times higher than that of other extraction methods.The correlation analysis showed that the antioxidant capacity of A.Radix extract had a marked positive correlation with the yield of total saponins,the content of astragaloside II,and maillard reaction.
