银耳多糖提取工艺的响应面法优化及抗氧化和保湿性研究

中文摘要: 以银耳为原料,在单因素试验基础上,采用响应面法优化银耳多糖提取工艺条件。结果表明,提取银耳多糖最佳工艺参数为液料比79∶1(mL/g)、提取温度91℃、提取时间5 h,此工艺条件下,测得多糖提取率为(20.29±0.15)%;银耳多糖对DPPH自由基、羟基自由基、ABTS+自由基清除能力呈剂量依赖性,在浓度为5 mg/mL时,3种自由基的清除率接近峰值,分别为47.31%、47.43%、52.14%,银耳多糖的抗氧化性是抗坏血酸的50%左右;在相对湿度43%、81%的环境下48 h,银耳多糖的吸湿率分别为25.15%、44.75%,远低于甘油的吸湿率(83.03%、122.65%),接近透明质酸钠(33.50%、53.50%);在干燥硅胶环境下48 h,银耳多糖的保湿率为42.15%,略低于透明质酸钠(49.50%)。

Abstract:The technological conditions of Tremella fuciformis polysaccharides were optimized by response surface methodology based on single factor experiment.The results showed that the optimum technological parameters of polysaccharides from T.fuciformis were as follows:liquid-to-material ratio of 79 ∶1(mL/g),extraction temperature of 91℃,and extraction time of 5 h.Under this condition,the extraction rate of polysaccharides was(20.29±0.15)%.The scavenging activity of T.fuciformis polysaccharides to DPPH,hydroxyl,and ABTS+free radicals was dose-dependent.When the concentration was 5 mg/mL,the scavenging rates of the three free radicals were 47.31%,47.43% and 52.14%,respectively.The antioxidant activity of T.fuciformis polysaccharides was about 50% of ascorbic acid.At 43% and 81% relative humidity for 48 h,the moisture absorption rate of T.fuciformis polysaccharides was 25.15% and 44.75%,respectively,which was much lower than that of glycerol(83.03% and 122.65%)and close to sodium hyaluronate(33.50% and 53.50%).Under dry silica gel condition for 48 h,the moisture retention rate of T.fuciformis polysaccharides was 42.15%,slightly lower than that of sodium hyaluronate(49.50%).
