玉米角质和粉质胚乳的异木聚糖结构及酶解特性的研究

中文摘要: 该文分别从玉米颗粒的角质胚乳和粉质胚乳中分离异木聚糖(heteroxylans,HX),通过形态粒度、理化特性、结构表征和酶解特性的测定与分析,研究不同玉米胚乳中异木聚糖的结构性质,进而揭示细胞壁降解酶在玉米酶解中的作用机理。通过结构分析可知,木聚糖主链只含有少量未取代的木糖残基(<9%),表明两者的侧链含量均较高。粉质胚乳中HX 比角质胚乳HX 含有更多的阿拉伯糖和半乳糖。角质胚乳中HX 的单基取代率更高(15%)。两种组分中糖醛酸的含量基本相当。粉质胚乳HX 中的半乳糖仅存在于末端位置。木聚糖酶制剂从粉质胚乳细胞壁中分离出的物质含量(40.8%)比角质胚乳(15.5%)多。此结果可能是由于两个组分之间存在结构差异,或受其结构对这些组分与其它细胞壁多糖的相互作用力的影响。
Abstract:In this paper,heteroxylans(HX)were isolated from the vitreous and floury endosperms of maize grains,respectively. Through the determination and analysis of morphological particle size,physicochemical properties,structural characterization,and enzymatic hydrolysis properties,the structural characterization of HX in different maize endosperms was studied,and the mechanism of cell wall in degrading enzymes during maize enzymatic hydrolysis was revealed.Structural analysis showed that the main chain of HX only contained a small amount of unsubstituted xylose residues(<9%),indicating that the content of both side chains was high.Compared with HX in the vitreous endosperm,HX in the floury endosperm contained more arabinose and galactose.The monobasic substitution rate of HX in the vitreous endosperm was higher(15%).The content of uronic acid in the two components was basically the same.Galactose of HX in the floury endosperm existed only in the terminal position. The material content of xylanase preparations isolated from the cell wall of the floury endosperm(40.8%)was higher than that of the vitreous endosperm(15.5%).The result was possibly due to the structural differences between the two components,or influenced by the structure on the interaction of the components with other cell wall polysaccharides.
