中文摘要: 为研究迷迭香提取物(rosemary extract,RE)对雌性果蝇延长寿命的分子机制,选取2 d 龄雌性果蝇饲养,将其随机分为4 组,分别为对照组、低剂量组(0.2 mg/mL RE)、中剂量组(0.5 mg/mL RE)、高剂量组(1.5 mg/mL RE),每3 d 更换一次培养基。在喂养不同时间点进行寿命、体重、摄食量、抗氧化酶活性、肠道解剖染色、实时定量聚合酶链式反应(quantitative real-time polymerase chain reaction,RT-qPCR)等实验。结果表明:添加RE 可延长13.10%寿命,且对氧化应激损伤有保护作用。RE 处理增强了内源性抗氧化酶活性,降低丙二醛(malondialdehyde,MDA)含量。此外,RE 抑制mTOR 的mRNA 表达,激活自噬以维持肠道稳态。同时,RE 改善肠道屏障功能,使肠道干细胞(intestinal stem cells,ISCs)异常增殖率降低35.89%,改善老年果蝇的肠道完整性。综上所述,RE 可通过胰岛素通路关键基因PI3K 和雷帕霉素途径关键基因mTOR 维持肠道稳态,提高抗氧化和应激抵抗能力,从而延长寿命。
Abstract:This study aimed to investigate the molecular mechanism of rosemary extract(RE)on the life extension of female Drosophila melanogaster.The 2-day-old female D.melanogaster were randomly divided into four groups:control group,low-dose group(0.2 mg/mL RE),medium-dose group(0.5 mg/mL RE)and highdose group(1.5 mg/mL RE)and the medium was changed every three days. Tests regarding lifespan,body weight,food intake and antioxidant enzyme activity as well as intestine anatomy and staining and quantitative real-time polymerase chain reaction(RT-qPCR)were performed on different days. The results showed that adding RE could prolong the lifespan by 13.10% and protect against oxidative stress injury. RE treatment enhanced the activity of endogenous antioxidant enzymes and decreased the content of malondialdehyde(MDA). In addition,RE inhibited the mRNA expression of mTOR and activated autophagy to maintain intestinal homeostasis.Moreover,RE improved the intestinal barrier function,reduced the abnormal proliferation rate of intestinal stem cells(ISCs)by 35.89%,and improved the intestinal integrity of elderly D.melanogaster.In conclusion,RE could maintain intestinal homeostasis through the key gene PI3K of insulin pathway and the key gene mTOR of rapamycin pathway,increase antioxidant and stress resistance abilities,and thus prolong life expectancy.
