竹虫蛋白酶解产物对肠道益生菌增殖作用的影响

中文摘要: 以竹虫为原料,考察其经3种水解酶(碱性蛋白酶、胰蛋白酶、木瓜蛋白酶)在不同酶解时间条件下酶解所制备的竹虫蛋白酶解液的益生活性。先对竹虫酶解液的蛋白回收率进行测定,然后将酶解液作为氮源替换益生菌培养基中的蛋白胨,通过监测4种肠道益生菌(植物乳杆菌LP45、乳双歧杆菌Probio-M8、鼠李糖乳杆菌GG、两歧双歧杆菌)生长曲线的变化,评价酶解液对益生菌增殖活性的调节作用。结果表明,各试验组菌悬液的OD600值普遍高于对照组,而代时普遍小于对照组;在相同酶解时间条件下,不同酶试验组之间菌悬液OD600值以及代时出现显著性差异(p<0.05),而同类酶不同酶解时间试验1组之间无显著性差异。同时,研究还发现竹虫蛋白酶解液对植物乳杆菌的促增殖效果最显著(代时增加率普遍小于60%),而经碱性蛋白酶制备的竹虫酶解液对肠道益生菌的效果最差(代时增加率普遍高于70%)。这表明,与对照组相比,竹虫蛋白酶解液均可在一定程度上促进4种益生菌增殖,且其活性与所用酶的种类密切相关,而与酶解时间无关。研究结果可为竹虫蛋白的益生活性研究及其在功能性食品中的开发利用奠定理论基础。

Abstract:Chilo Frusciden-talis Hampson was used as raw material to be hydrolyzed by three kinds of hydrolases(alkaline protease,trypsin,and papain)under different hydrolytic time to prepare the protease hydrolysates,and then analyzed the probiotic activities.The protein recovery rates of different protease hydrolysates were determined,and the protease hydrolysates were used as nitrogen source to replace peptone in the probiotic culture medium of four kinds of intestinal probiotics including Lactobacillus plantarum LP45,Bifidobacterium lactobacillus Probio-M8,Lactobacillus rhamnoides GG,and Bifidobacterium bifidobacteria.Through monitoring the growth curves of intestinal probiotics,the regulatory effect of protease hydrolysates on the proliferation activity of probiotics was evaluated.The results showed that the OD600value of the experimental intervention group was generally higher than that of the control group,and the generation time was generally less than that of the control group.Under the same hydrolytic time,the OD600value and generation time of the experimental intervention groups with different enzyme types showed significant differences(p<0.05),but that of the experimental intervention group 1 with the homologous enzyme under different hydrolytic time showed no significant difference.At the same time,the study also found that protease hydrolysates of C.Frusciden-talis Hampson had the most obvious effect on the replication and proliferation of Lactobacillus plantarum(the increase rate of generation time was generally less than 60%),whereas protease hydrolysates of C.Frusciden-talis Hampson prepared by alka-line protease had the worst effect on intestinal probiotics(the increase rate of generation time was generally higher than 70%).The results showed that compared with the control group,protease hydrolysates of C.Frusciden-talis Hampson promoted the proliferation of four probiotics to some extent,and the activities were related to the enzyme types rather than the hydrolytic time.The findings of this study provided references for the study on the probiotic activities of C.Frusciden-talis Hampson and the development and utilization of C.Frusciden-talis Hampson in functional food.
