中文摘要: 该研究采用80%乙醇溶液对玉米须进行提取,进一步利用不同极性溶剂进行萃取,采用液相色谱串联质谱法对玉米须乙醇提取物中的正丁醇萃取物进行活性成分分析,并研究其抗氧化活性、消化酶抑制活性以及其对链脲佐菌素诱导的2型糖尿病小鼠的降血糖作用。结果表明,与石油醚萃取物和乙酸乙酯萃取物相比,正丁醇萃取物中总多酚、总黄酮的含量最高,抗氧化活性最强,具有一定的消化酶抑制活性。正丁醇萃取物可以缓解2型糖尿病小鼠体质量的下降趋势,降低血糖和血清胰岛素水平,改善葡萄糖耐受性,调节血脂水平,提高抗氧化酶活性。组织切片病理学结果显示,正丁醇萃取物具有细胞保护作用。综上,玉米须乙醇提取物中的正丁醇萃取物具有良好的降血糖功效。
Abstract:In this study,80% ethanol solution was used to extract corn silk,and then different polar solvents were used to further extract.The active components of n-butanol fraction(NBF)from corn silk ethanol extract were analyzed by liquid chromatography tandem mass spectrometry(LC-MS/MS)method.The antioxidant activity,digestive enzyme inhibitory activity,and hypoglycemic effect on streptozotocin-induced type 2 diabetic mellitus mice were studied.The results showed that NBF had the highest content of total polyphenols and total flavonoids compared with petroleum ether and ethyl acetate fractions,with the strongest antioxidant activity and certain digestive enzyme inhibitory activity.NBF alleviated the declining trend of the weight of type 2 diabetic mellitus mice,and reduced blood glucose and serum insulin levels.NBF improved glucose tolerance,regulated blood lipid level,and improved the activities of antioxidant enzymes.Histopathological results showed that NBF had cytoprotective effects.Therefore,NBF from corn silk ethanol extract had good hypoglycemic effect.
