铁皮石斛花多糖的复合酶解法提取及生物有效性研究

中文摘要: 研究一种新型有效的铁皮石斛花多糖提取方法,并对铁皮石斛花多糖、黄酮和多酚的生物有效性进行测定,为充分开发利用铁皮石斛资源提供科学依据。采用复合酶解法对铁皮石斛花进行提取,用蒽酮-硫酸法测定铁皮石斛花多糖含量,以热水浸提法为对照,评价铁皮石斛花复合酶解法的生物有效性。试验结果得到复合酶最佳配比:酶用量为纤维素酶添加量0.8%,果胶酶添加量0.4%,碱性蛋白酶添加量0.4%;酶解提取工艺最优参数为料液比1∶20(g/mL),酶解pH4,酶解时间2.0 h,酶解温度40℃;提取液的生物有效性为多糖生物利用度32.61%、黄酮生物利用度22.34%、多酚生物利用度19.18%。复合酶解法提取铁皮石斛花,提取液多糖含量较高,提取率为169.3 mg/g,可较好地维持多糖、黄酮、多酚的生物有效性。
Abstract:This study aims to explore a new and effective method for extracting polysaccharides from Dendrobium officinale flower and determine the bioavailability of the extracted polysaccharides,flavonoids,and polyphenols,which is expected to lay a scientific basis for the further development and utilization of D.officinale resources.To be specific,compound enzymatic hydrolysis was used to extract the flower of D.officinale,and the content of polysaccharides in the flower of D.officinale was determined with anthrone-sulfuric acid method.With hot water extraction method as a reference,biological effectiveness of the compound enzyme hydrolysis was evaluated.The results showed that the optimal ratio of compound enzymatic was as follows:0.8% of cellulase,0.4% of pectinase,0.4% of alkaline protease,material-to-liquid ratio of 1∶20(g/mL),hydrolysis at pH4 and 40 ℃for 2.0 h.The bioavailability of the yielded polysaccharides,flavonoids,and polyphenols was 32.61%,22.34%,and 19.18%,respectively.The content of polysaccharides in the extract of D.officinale flower by compound enzyme hydrolysis was high,and the extraction rate was 169.3 mg/g.
