中文摘要: 为研究低压静电场处理对采后马铃薯块茎愈伤效果的影响,以“冀张薯12号”马铃薯块茎为试材,人工模拟机械损伤后,置于有低压静电场的常温[(20±2)℃]、相对湿度85%~90%、黑暗条件下愈伤,通过测定块茎愈伤期间的失重率、损伤伤口处硬度、褐变度、苯丙烷代谢关键酶活性和代谢产物含量的变化,以及分析低压静电场对过氧化物酶、多酚氧化酶活性和H2O2含量的影响来评价愈伤效果。结果表明,在愈伤期间低压静电场处理能够有效减少损伤块茎的重量损失,显著促进损伤部位褐变度值和硬度的增加,显著提高伤口处组织中苯丙氨酸解氨酶活性,促使总酚、类黄酮和木质素含量增加,此外,低压静电场处理增加了块茎伤口处的多酚氧化酶、过氧化物酶活性和H2O2含量,结果显示,低压静电场处理能够促进采后马铃薯块茎的愈伤。
Abstract:The effects of low-voltage electrostatic field treatment on the healing effect of post-harvest potato tubers were investigated in'Jizhangshu No.12'potato tubers,which served as the test material.After artificial mechanical damage,the calluses were placed under normal temperature(20±2)℃ and relative humidity(85%-90%)conditions in the dark and subjected to a low-voltage electrostatic field.Changes in weight-loss rate,hardness of injury wounds,browning degree,activity of key enzymes associated with phenylpropane metabolism,and contents of metabolites were measured.The effects of the low-voltage electrostatic field on peroxidase,polyphenol oxidase activity,and hydrogen peroxide (H2O2)contents were analyzed to evaluate the effects of the electrostatic field on the calluses.During the callus period,the low-voltage electrostatic field treatment effectively reduced the weight loss of damaged tubers and significantly promoted increase of the browning degree and hardness of the damaged parts.After the low-voltage electrostatic field treatment,the activity of phenylalanine ammonia lyase in the tuber wounds increased significantly and the contents of total phenols,flavonoids,and lignin also increased.In addition,the low-voltage electrostatic field treatment also increased the activities of polyphenol oxidase and peroxidase and H2O2contents in the tuber wounds.These results showed that low-voltage electrostatic field treatment can promote callus formation in postharvest potato tubers.
