中文摘要: 为优化金花茶叶多酚的提取工艺及分析其抗氧化活性,该试验采用超声波辅助法提取金花茶叶多酚,考察乙醇浓度、提取时间、提取温度和料液比对金花茶叶多酚提取量的影响,并通过正交试验设计对提取工艺进行优化。以DPPH、ABTS+自由基清除率和总抗氧化能力为指标,评价金花茶叶多酚提取物的体外抗氧化活性。结果表明,金花茶叶多酚的最佳提取工艺参数组合为乙醇浓度90%、提取时间30 min、提取温度35℃、料液比1∶50(g/mL)。在此条件下,多酚提取量达到295.82 mg/g。抗氧化活性试验表明,金花茶叶多酚具有较好的总抗氧化能力,对DPPH和ABTS+自由基具有较强的清除作用,其IC50值分别为0.030 9、0.043 4 mg/mL,同时总抗氧化能力为0.82 mmol/L。研究表明金花茶叶多酚具有作为天然抗氧化剂的潜力。
Abstract:The present study aimed to optimize the extraction process of polyphenols from Camellia petelotii(Merrill)Sealy and analyze their antioxidant activity.An ultrasonic-assisted extraction method was used to extract polyphenols from Camellia petelotii leaves.The effects of ethanol concentration,extraction time,extraction temperature and ratio of material to liquid on the extraction yield of polyphenols were investigated.In addition,the extraction process was optimized based on orthogonal experimental design.The scavenging rates of 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)free radicals,and total antioxidant capacity were used as indices to evaluate the antioxidant activity of polyphenolic extracts from Camellia petelotii leaves in vitro.The results showed that the optimal extraction parameters of polyphenols from Camellia petelotii leaves were an ethanol concentration of 90%,extraction time of 30 min,extraction temperature of 35 ℃,and material-liquid ratio of 1∶50(g/mL),which achieved a polyphenol extraction yield of 295.82 mg/g.The antioxidant activity results showed that polyphenols from Camellia petelotii leaves exhibited good total antioxidant capacity and a strong scavenging effect on DPPH and ABTS+free radicals,and their IC50values were 0.030 9 mg/mL and 0.043 4 mg/mL,respectively;the total antioxidant capacity was 0.82 mmol/L.According to the results,polyphenols from Camellia petelotii leaves could be used as natural antioxidants.
