中文摘要: 从丽江山荆子中提取出表儿茶素单体,并对其体外抗氧化能力进行研究。首先采用甲醇浸提、超声波辅助提取,再分别经MCI柱层析、ODS柱层析、Sephadex柱层析分离纯化。采用高效液相色谱法分析表儿茶素纯度,采用核磁共振(nuclear magnetic resonance,NMR)以及电喷雾电离-串联质谱法(electrospray ionization-tandem mass spectrometry,ESI-MS)确定化学结构。用VC作对照检测表儿茶素对DPPH自由基,ABTS+自由基,羟基自由基的清除能力及其还原能力的测定。经一系列分离纯化操作后,测得其纯度为98%。经分离纯化后的表儿茶素均能清除DPPH自由基、ABTS+自由基、羟基自由基以及具有一定的还原能力。
Abstract:The antioxidant activity of epigallocatechin,extracted and purified from Malus rockii Rehd.,was measured in vitro.Epigallocatechin was extracted by methanol and ultrasound assisted,and purified using MCI gel,octadecylsilyl,and Sephadex column chromatography.Epigallocatechin purity was evaluated using high-performance liquid chromatography and nuclear magnetic resonance,and its chemical structure was determined using electrospray ionization-mass spectrometry.The ability of epigallocatechin to scavenge free radicals from DPPH,ABTS+,and hydroxyl,as well as its reductive capacity,were evaluated using VCas a positive control.The purity of the extracted epigallocatechin was 98%,epigallocatechin exhibited a strong ability to scavenge DPPH,ABTS+,and hydroxyl-derived free radicals,as well as a more specific term reduction.
