酸浆果实多酚和多糖双水相提取工艺及其抗氧化活性

中文摘要: 为优化酸浆果实中多酚和多糖双水相提取工艺,并初步评价其体外抗氧化活性,采用超声辅助C2H5OHK2HPO4双水相萃取酸浆果实多酚和多糖,以酸浆果实多酚、多糖萃取率为指标,依据单因素试验设计正交试验优化粗提工艺;利用1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-trinitrophenylhydrazine,DPPH)、铁离子还原能力(ferric ion reducing antioxidant power,FRAP)法评价酸浆果实多酚和多糖体外抗氧化活性。结果表明:在最优提取条件[温度20℃,料液比1∶40(g/mL),乙醇质量分数24%、K2HPO4质量分数20%]下,酸浆多酚萃取率为83.44%;在最优工艺[温度为55℃,料液比1∶40(g/mL),乙醇质量分数18%,K2HPO4质量分41%数]下,酸浆多糖萃取率为89.16%;抗氧化实验显示,酸浆多酚浓度10 mg/mL时DPPH自由基清除能力为93.21%,铁离子还原能力为0.269 6 mg/mL,均强于酸浆多糖。该研究表明酸浆多酚、多糖均具有一定的抗氧化能力;超声辅助双水相同时提取酸浆果实多酚、多糖方法是可行的。

Abstract:The present study aimed to optimize the extraction conditions of polyphenols and polysaccharides from Physalis alkekengi fruit using aqueous two-phase ultrasonic-assisted methodology and evaluate the antioxidant activity of the extract.Ultrasound was used to assist C2H5OH-K2HPO4aqueous two-phase extraction of polyphenols and polysaccharides from P.alkekengi fruit.The extraction rates were used as indicators.The crude extraction process was optimized using a single factor design orthogonal experiment.To evaluate the antioxidant activity of polyphenols and polysaccharides from P.alkekengi fruit,the 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)and ferric reducing antioxidant power methods were applied.Under optimal extraction conditions for polyphenols,an aqueous two-phase system was constructed with a temperature of 20 °C,material-to-liquid ratio of 1∶40(g/mL),C2H5OH mass fraction of 24%,and K2HPO4mass fraction of 20%.The extraction rate of P.alkekengi fruit polyphenols was 83.44%.For the extraction of polysaccharides,an aqueous two-phase system was constructed with a temperature of 55 °C,material-to-liquid ratio of 1∶40(g/mL),C2H5OH mass fraction of 18%,and K2HPO4mass fraction of 41%.The polysaccharide of P.alkekengi fruit extraction rate was 89.10%.The results of these antioxidant experiments showed that the ability of P.alkekengi fruit polyphenols to scavenge DPPH free radicals and reduce ferric ions were stronger than those of P.alkekengi fruit polysaccharides.Thus,polyphenols and polysaccharides from P.alkekengi fruit have certain antioxidant capacities.Therefore,optimizing the extraction method using the ultrasonic-assisted double water method is promising and feasible.
