中文摘要: 为改善黑曲霉发酵后期产糖化酶速率,通过二氧化碳生成速率(carbon evolution rate,CER)指导补料,对发酵后期的补料方式进行优化。结果表明,在发酵中后期CER开始出现下降趋势并持续36 h后停止补料,直至CER反弹为峰值时恢复补料能够明显提升氧消耗速率,糖化酶在发酵后期依然保持较高的合成速率,最终糖化酶酶活性达到20 895 AGI/mL,比优化之前提高了60%。
Abstract:In order to improve the enzymatic saccharification rate in the later stage of Aspergillus niger fermentation,the dosing mode was optimized by using the carbon evolution rate(CER).The results showed that by stopped doing when CER started to dropped for 36 h and recovered dosing when the CER rebound back to the peak in middle and later period of fermentationcan,the oxygen consumption rate was significantly improved,which maintained the glycosylase synthesis high at the later stage of fermentation.The final glucoamylase enzymatic activity reached 20 895 AGI/mL,increasing by 60%compared with that of before optimization.
