中文摘要: 为研究巨大芽孢杆菌(Bacillus megaterium)LB01粗提物对采后芒果炭疽病的防治效果及粗提物活性成分的分离鉴定,采用乙酸乙酯、石油醚和二氯甲烷等有机溶剂对此菌株LB01发酵滤液进行萃取获得粗提物,通过平板扩散法和芒果活体实验法测定巨大芽孢杆菌LB01粗提物对采后芒果炭疽病菌的体外及体内抗菌活性,采用多种色谱柱对粗提物进行分离,运用核磁共振技术结合文献数据鉴定结构。结果表明,乙酸乙酯和石油醚粗提物抑制平板上病原菌生长的效果优于二氯甲烷。乙酸乙酯和石油醚粗提物在培养2 d后对病原菌生长的抑制率最高,分别为(76.2±5.2)%和(75.3±6.5)%。乙酸乙酯粗提物(200 μL、80 μg/mL)对采后芒果炭疽病的体内防治效果与化学杀菌剂施保克相似,7 d后生防效果高达(89.55±1.12)%,此粗提物共分离出3种化合物,分别为4-羟基苯乙酸、4-氨基苯甲酸、3-甲氧基-4-羟基肉桂酸。
Abstract:Ethyl acetate,petroleum ether,and dichloromethane extracts of the fermentation filtrate of Bacillus megaterium LB01 were used to study the biocontrol effect of B.megaterium LB01 crude extract on postharvest mango anthracnose and structure of active components.The in vitro and in vivo antifungal activities of the Bacillus megaterium LB01 crude extract against Colletotrichum gloeosporioides were determined by plate diffusion method and mango in vivo experiment.The crude extract was separated by column chromatography,and the structures of the constituents were identified by nuclear magnetic resonance and literature data.The results showed that the inhibition effect of ethyl acetate and petroleum ether extracts was better than that of dichloromethane.Ethyl acetate(76.2±5.2)% and petroleum ether(75.3±6.5)% extracts had the highest inhibition rate on the growth of pathogens after two days of culture.The control effect of ethyl acetate extract(200 μL,80 μg/mL)on mango anthracnose in vivo was similar to that of the chemical fungicide sportak.After seven days,the biocontrol effect was up to(89.55±1.12)%.Three compounds were separated from the crude extract,namely 4-hydroxyphenylacetic acid,4-aminobenzoic acid,and 3-methoxy-4-hydroxycinnamic acid.
