中文摘要: 百香果(Passiflora edulia Sims)风味浓郁且果实多汁,素有“果汁之王”的美誉。但不同栽培品种间甜味差异显著,导致风味多样,决定水果甜味和风味的主要因素是糖含量。为深入研究百香果糖含量与糖代谢相关基因的关系,通过气相色谱-质谱分析(gas chromatography-mass spectrometry,GC-MS)对3个不同品种(紫香、黄金和满天星)的百香果果实糖含量进行分析。从百香果转录组注释数据中获得糖代谢密切相关的基因,并对其表达量进行分析。结果表明:黄金百香果葡萄糖、D-果糖、蔗糖的平均含量分别为238.0、228.7、91.0 mg/g,满天星葡萄糖、D-果糖、蔗糖的平均含量分别为 229.7、226.3、55.6 mg/g,而紫香葡萄糖、D-果糖、蔗糖的平均含量分别为 218.3、210.0、86.4 mg/g。通过实时荧光定量多聚核苷酸链式反应技术(real-time quantitative polymerase chain reaction,RT-qPCR)对糖代谢相关基因:磷酸烯醇丙酮酸羧化激酶、磷酸果糖激酶、丙酮酸脱羧酶在3个品种百香果中的表达量进行分析,发现编码磷烯醇丙酮酸羧化与丙酮酸脱羟酶的两个基因在不同品种间表达量存在显著差异。
Abstract:Passion fruit has a strong flavor.However,there can be sharp distinctions between the sweet tastes of different cultivars.The main factor determining a cultivar's sweetness and flavor is its sugar content.To characterize relationships between sugar content and expression of sugar metabolism-related genes in three varieties of passion fruit,a qualitative and quantitative analysis of sugar content has been carried out by gas chromatography-mass spectrometry (GC-MS).Genes that are closely related to sugar metabolism were obtained from the annotation data of the passion fruit transcriptome,and expression levels were analyzed.The Gold cultivar displayed average glucose,fructose,and sucrose contents of 238.0,228.7,91.0 mg/g,respectively.The Mantianxing cultivar contained 229.7,226.3,55.6 mg/g,respectively.The Purple cultivar contained 218.3,210.0,86.4 mg/g,respectively.RT-qPCR was used to detect phosphoenolpyruvate carboxylation kinase,phospho fructose kinase,and pyruvate decarboxylase levels.It was found that the expression levels of the two genes encoding phosphoenolpyruvate carboxyl kinase and pyruvate decarboxylase were significantly different among different varieties.
