产正丁醇脱氢酶微生物的筛选及其酶学性质的初步研究

中文摘要: 为研究酒类混合发酵过程中正丁醇的产生规律及建立调控正丁醇含量的方法,以酒曲和酒醅为样品,利用LB培养基分离菌株,吸光值法和气相色谱法测定菌株对正丁醇催化的减少量,从中筛选能够在乙醇存在条件下催化正丁醇的微生物,并对该微生物所产酶的性质进行初步研究。结果显示,所筛得编号为DQ-54的细菌属肠杆菌属(Enterobacter sp.),其产生的酶(Ec-54ADH)能够在含有5.00%乙醇溶液中转化正丁醇为正丁醛,且随着乙醇浓度的增加(0%~25%),Ec-54ADH对正丁醇的特异性改变,催化能力显著降低(p<0.05),但对乙醇的催化能力在乙醇浓度为5.00%~15.00%时没有显著差异(p>0.05)。另外,在乙醇存在条件下(3.00%),当pH值为7.0和温度为40℃时,Ec-54ADH具有最高催化正丁醇活性,添加金属离子(Fe2+、Mg2+、Mn2+、Zn2+和Cu2+)能够抑制Ec-54ADH对正丁醇催化活力。
Abstract:In order to reveal the rule of producing n-butanol in the process of mixed fermentation of alcohol beverage and establish the method to control the content of n-butanol of alcohol beverage,the plate separation,microplate spectrophotometer and gas chromatography were applied to screen the ethanol-tolerance butanol dehydrogenase(Ec-54ADH)that could catalyze n-butanol in the presence of ethanol.The results showed that the isolate DQ-54 was belong to Enterobacter sp.and Ec-54ADH from DQ-54 could convert n-butanol to butyraldehyde in 5.00% ethanol.However,the activity of catalyzing n-butanol of Ec-54ADH was decreased significantly(p<0.05)with the increase of the ethanol concentration(0%-25%)and the activity of catalyzing ethanol of Ec-54ADH was no significant difference(p>0.05)in the 5.00%-15.00% of ethanol concentration.In addition,Ec-54ADH had the highest catalytic activity when pH was 7 and the temperature was 40℃ and the addition of metal ions (Fe2+,Mg2+,Mn2+,Zn2+and Cu2+)could inhibit the catalytic activity of Ec-54ADH to nbutanol under the condition of 3.00% ethanol.
