浓香型白酒窖泥微生物群落多样性及理化因素对其影响

中文摘要: 采用Illumina Miseq高通量测序技术解析豫酒某浓香型酒企4年和40年窖池的窖壁、窖底泥原核微生物群落多样性和组成。结果表明,窖泥样品操作分类单元(operational taxonomic units,OTU)数量、Chao1指数和ACE指数没有显著性(P>0.05)差异,40年窖底泥香农(Shannon)指数和辛普森(Simpson)指数显著性(P<0.05)偏低。窖泥优势菌门为厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、广古菌门(Euryarchaeota)和放线菌门(Actinobacteria);优势菌属为乳酸杆菌属(Lactobacillus)、产己酸菌属(Caproiciproducens)、不动菌属(Acinetobacter)、理研菌属(Petrimonas)等共12个菌属,隶属于4个门,且集中在厚壁菌门(Firmicutes)。厚壁菌门(Firmicutes)在40年窖底泥中占绝对优势,Bacteroidetes和Euryarchaeota在4年窖底泥和40年窖壁泥中含量都较高;Lactobacillus为40年窖壁、窖底泥第一大优势菌属,而4年窖壁泥和窖底泥第一优势菌属分别为Acinetobacter和Caproiciproducens。冗余分析(redundancy analysis,RDA)显示,总酸(30.7%)和pH值(15.7%)与窖泥优势菌群的相关性最高且显著(P<0.05),说明总酸和pH值可能是影响上述不同类型窖泥优势菌群空间分布的主要理化因子。
Abstract:In order to clarify the structure of microbial community in pit mud of one Luzhou-flavor liquor of Yujiu enterprise,Illumina Miseq high-throughput sequencing technology was used to analyze the diversity and composition of prokaryotic microbial communities in the walls and bottom mud for 4 years and 40 years.The results showed that there was no significant difference(P>0.05)between observed operational taxonomic units(OTU),Chao1 and ACE of pit mud,and the Shannon and Simpson of 40 years pit bottom mud had significant significance(P<0.05).The dominant phylum of the pit mud was Firmicutes,Proteobacteria,Bacteroidetes,Euryarchaeota and Actinobacteria.There were 12 dominant genus.The dominant genus was Lactobacillus,Caproiciproducens,Acinetobacter,Petrimonas,etc,belonging to 4 phylum,and mainly concentrated Firmicutes.Firmicutes had an absolute advantage in the pit bottom mud for 40 years,Bacteroidetes and Eur-yarchaeota had higher contents in the pit bottom mud for 4 years and the pit wall mud for 40 years.Lactobacillus was the first dominant bacterial genera of wall and bottom pit mud for 40 years.While the first dominant bacteria of wall and bottom pit mud for 4 years were Acinetobacter and Caproiciproducens,respectively.Redundancy analysis(RDA)showed that the total acid(30.7%)and pH (15.7%)had the highest correlation with the dominant microbial community in the pit mud (P<0.05),indicating that total acid and pH were the dominant physicochemical factors affecting the distribution of dominant bacteria in different types pit mud.
