黄绿蜜环菌硒多糖抗氧化活性及抑菌活性研究

中文摘要: 为研究黄绿蜜环菌硒多糖的抗氧化活性及抑菌活性,采用乙醇沉淀法、Sevag法从黄绿蜜环菌富硒培养液中获得硒多糖,体外测定其对羟基自由基、DPPH自由基和ABTS+自由基的清除能力,并采用滤纸片扩散法测定对大肠杆菌和金黄色葡萄球菌的抑制作用。经测定,黄绿蜜环菌硒多糖中硒和多糖含量分别为(2.84±0.23)g/kg和(56.27±0.35)%。研究结果表明,硒多糖对羟基自由基、DPPH自由基、ABTS+自由基的清除能力随着其浓度的升高而显著增加(P<0.05),但清除率均小于抗坏血酸。高浓度的黄绿蜜环菌硒多糖对大肠杆菌(Escherichia coli)生长的抑制效果明显高于低浓度(P<0.05),但对金黄色葡萄球菌(Staphylococcus aureus)无抑制作用。综上所述,黄绿蜜环菌硒多糖具有一定的抗氧化活性和抑菌效果,其生物活性强弱与浓度有关。
Abstract:In order to investigate antioxidant and antimicrobial activities of selenium-containing polysaccharide extracted from Armillaria luteo-virens,selenium-containing polysaccharide were partitioned from Se-enriched fermentation by ethanol precipitation method and Sevag method in turn,and evaluated in vitro the·OH,DPPH·and ABTS+·scavenging activity,respectively,and assessed antimicrobial activity using agar diffusion method.The results showed that the concentrations of selenium and polysaccharide in selenium-containing polysaccharide were(2.84±0.23)g/kg and(56.27±0.35)%,respectively.The significantly enhancing removal of·OH,DPPH·and ABTS+·scavenging activity in vitro were observed with the increasing selenium-containing polysaccharide(P<0.05).However the DPPH,hydroxyl and ABTS+radical scavenging potential of seleniumcontaining polysaccharide were lower than ascorbic acid.In addition,The high concentrations of seleniumcontaining polysaccharide inhibited significantly the growth of Escherichia coli compared to the low concentrations (P <0.05),without the inhibitory effect on Staphylococcus aureus.Selenium-containing polysaccharide from A.luteo-virens had certain antioxidant and antimicrobial activities.There were some correlations between activities and concentrations.
